Abstract/Summary:
Objective:To study and investigate the expression and clinical significance of Lgr5(Leucine-rich repeat sequence G protein-coupled receptor)and VEGF(vascular endothelial growth factor)in cervical cancer.Methods:Paraffin sections of 109 gynecological surgical excision and biopsy specimens were collected from Zhucheng People’s Hospital and Zhucheng Maternal and Child Health Hospital from January 2010 to December 2015.The age distribution of the patients ranged from 27 to 71 years.The selected pathological wax block tissues in this study included 12 cases of normal cervical tissues,20 cases of low-grade cervical tissues,20 cases of high-grade squamous intraepithelial lesion tissues,and 57 cases of squamous cervical carcinoma tissues.Due to the small number of surgical pathology cases of adenocarcinoma of the uterine cervix,they were not included in this study.Among them,cervical squamous carcinoma tissues were classified according to pathological differentiation: 31 cases in the low differentiation group(grade III),20 cases in the medium differentiation group(grade II),and 6 cases in the high differentiation group(grade I);classified according to clinical stage of cervical cancer(FIGO,2018): 30 cases in stage I,17 cases in stage IIa,and 10 cases in stage IIIc1p(lymph node metastasis was found when the suspected lesion tissues were taken for pathological diagnosis intraoperatively);classified according to whether lymph node metastasis was found in squamous cervical cancer tissues were classified as: 10 cases with lymph node metastasis were found and 47 cases without lymph node metastasis;according to the size of cervical cancer tumor lesions were classified as: tumor < 2cm: 34 cases,2cm ≤ tumor < 4cm: 15 cases,and tumor ≥ 4cm: 8 cases.The expression of Lgr5 and VEGF was observed mainly by staining in their tissues using immunohistochemical methods,and SPSS 20.0 software was selected for statistical analysis and test scoring to study and analyze the positive expression of Lgr5 and VEGF in different groups of cervical cancer tissues and the significance of clinical guidance.Results:1.The positive expression rates of Lgr5 in normal cervical,low and high grade squamous intraepithelial lesions,and cervical squamous carcinoma tissues were: 8.33%(1/12),20%(4/20),65%(13/20),and 73.68%(42/57),respectively,with a significant difference(P<0.05).The positive rates in the ages of 27-49 and 50-71 years were 76.92%(20/26)and 70.97%(22/31),respectively,with a non-significant difference(P>0.05).The positive rates in the absence and presence of lymph node metastasis were: 68.09%(32/47)and100%(10/10),respectively,with a significant difference(P < 0.05).The positive rates in high,medium and low-differentiated carcinomas were: 16.67%(1/6),65%(13/20)and90.32%(28/31),respectively,with significant differences(P<0.05).The positive rates in different tumor lesions were: tumor <2cm: 61.76%(21/34),2cm≤ tumor <4cm: 86.67%(13/15),and tumor ≥4cm: 100%(8/8),respectively,with significant differences(P<0.05).The positive rates in stage I,IIa,and IIIc1 p were 66.67%(20/30),70.59%(12/17),and100%(10/10),respectively,with statistically insignificant differences(P > 0.05).2.The positive expression rates of VEGF in normal cervical,low and high grade squamous intraepithelial lesions,and squamous cervical cancer tissues were: 8.33%(1/12),15%(3/20),25%(5/20),and 56.14%(32/57),respectively,with a significant difference(P<0.05).The positive rates in the ages of 27-49 and 50-71 years were 57.69%(15/26)and 54.84%(17/31),respectively,with a non-significant difference(P>0.05).The positive rates in high,medium and low-differentiated cancers were: 33.33%(2/6),40%(8/20)and 70.96%(22/31),respectively,with a significant difference(P<0.05).The positive rates in the absence and presence of lymph node metastasis were: 46.81%(22/47),100%(10/10),respectively,with a significant difference(P<0.05).The positive rates in stage I,IIa and IIIc1 p were 36.67%(11/30),64.71%(11/17)and 100%(10/10),respectively,with significant differences(P<0.05).The positive rates in different tumor lesions were: tumor <2cm: 35.29%(12/34),2cm≤ tumor <4cm: 80%(12/15),and tumor≥4cm: 100%(8/8),respectively,with significant differences(P<0.05).3.Among the 57 cervical cancer tissue specimens participating in the test,there were 30 cases in which both VEGF and Lgr5 were positively expressed,13 cases in which Lgr5 was positively expressed and VEGF was negatively expressed,and 2 cases in which Lgr5 was negatively expressed and VEGF was positively expressed.Correlation analysis using Spearman correlation coefficient showed a positive correlation between VEGF and positive expression of Lgr5(r=0.516,P<0.001).Conclusions:1.The expression of Lgr5 and VEGF were enhanced in different cervical lesion tissues in different degrees.2.Positive expression of VEGF was significantly correlated with the presence of lymph node metastasis,degree of pathological differentiation,size of tumor lesions,and clinical stage of cervical cancer tissues confirmed(P<0.05),but not with age(P>0.05).positive expression of Lgr5 was significantly correlated with the presence of lymph node metastasis,degree of pathological differentiation,and size of tumor of cervical cancer tissues confirmed(P<0.05),but no correlation with clinical stage and age(P>0.05).3.There is a relationship between the expression of Lgr5 and VEGF in cervical cancer tissues,and the two may play a synergistic role in the developmental process of cervical cancer.
