Objective:To test the expression of Lgr5,CD44v6and COX-2in human gastriccancer,and to analyze the correlation with clinicopathologic features and potentialapplications for clinical diagnosis and treatment.Methods:Using immunohistochemistry,Lgr5,CD44v6,COX-2and Ki-67were analyzedin41RO-resected primary gastric adenocarcinomas,while paracancerous tissues were alsocollected as control from20concurrent patients.Results:1.The expression rates of Lgr5,CD44v6,COX-2and Ki-67in gastric carcinoma weresignificantly higher than that in paracancerous tissues(P<0.05).Moreover,the spatialdistribution of these targets changed.2.Lgr5expression was less frequent in cancers with advanced TNM-stage or poordifferentiation(P<0.05).On the contrary,the others were positively correlated with thepresence of the metastases in the regional lymph nodes and the advanced pTNMstage(P<0.05).The COX-2immunoreactions have also turned positive more frequently inadvanced T-stage cancer(P<0.05).There was no significant difference inLgr5,CD44v6,COX-2and Ki-67based on the age,sex and Lauren classification.3. A positive correlation between Ki-67and CD44v6expression was observed in gastricancer tissues(P<0.05).Conclusions:1. Lgr5,CD44v6, COX-2and Ki-67may play an important role in gastric carcinogenesisand progression.2. The expression of Lgr5was down-regulated in most high malignant degree andinvasive gastric cancer tissues.3. CD44v6,combined with Ki-67,would be powerful markers to predict the metastases in the regional lymph nodes and be usefull to conduct therapy.4. Lgr5,CD44v6and COX-2could be potential targets for treatment of gastric cancer.
