Objective: The proportion of obese patients in type 2 diabetes is high.Controlling the weight of obese diabetic patients can help relieve their blood sugar and blood lipid levels.There have been many studies on the regulation of glucose metabolism by electroacupuncture in diabetic mice,but there are few studies on the regulation of lipid metabolism by electroacupuncture.The specific mechanism remains to be further explored.This study intends to establish a mouse model of type 2 diabetes(T2DM)to observe the effects of electroacupuncture on food intake,body weight,epididymal fat mass,triglycerides,cholesterol,high-density lipoprotein,and lowdensity lipoprotein.And whether electroacupuncture regulates triglyceride levels in diabetic mice via PPAR-γ/UCP2-AMPK-ACC signaling pathway of the liver,so as to explore the possible mechanism of electroacupuncture regulating the levels of triglyceride in type 2 diabetic mice.Methods: Mice were induced to type 2 diabetes by high-fat diet and STZ intraperitoneal injection,and were divided into 4 groups randomly: control group,diabetic electroacupuncture stimulation group(DM+EA group),type 2 diabetes group(DM group),diabetic pseudo electroacupuncture stimulation group(DM+SEA group).After successfully establishing the type 2 diabetes model,electroacupuncture stimulation was performed for 8 weeks.At 1,3,5,6,7,and 8 weeks,the body weight and 24-hour food consumption of the mice were monitored.Eight weeks later,the weight of epididymal fat,the levels of triglycerides,cholesterol,high-density lipoprotein,and low-density lipoprotein in each group of the mice were evaluated;western blot and q RT-PCR were used to detect PPAR-γ,UCP2,p AMPK,p ACC m RNA and protein level in liver tissue of mice.Results:(1)Compared with the control group,the weight gain of mice in the DM group decreased,and the fat mass of the epididymis increased significantly.Compared with the DM group,the 24-hour food consumption,body weight and epididymal fat mass of the DM+EA group were significantly reduced.(2)Compared with the control group,the contents of triglyceride,cholesterol,high-density lipoprotein and lowdensity lipoprotein of the DM group increased,but there was no statistically significant difference between the two groups of the high-density lipoproteins.Compared with the DM group,the electroacupuncture stimulation can reduce the triglyceride level of diabetic mice,the difference is statistically significant,but has no significant effect on the levels of cholesterol,high density lipoprotein and low-density lipoprotein.(3)Compared with the control group,the m RNA and protein expression of PPAR-γ and UCP2 in the DM group increased,and the protein expression of p AMPK and p ACC decreased.Compared with the DM group,the electroacupuncture stimulation reduced the m RNA and protein expression of PPAR-γ,and increased the m RNA and protein expression of UCP2 and the protein expression of p AMPK and p ACC in the DM+EA group.Conclusion: Electroacupuncture can reduce the appetite of type 2 diabetic mice,reduce their food intake,body weight and epididymal fat mass,and improve the level of triglycerides in type 2 diabetic mice by regulating the PPAR-γ/UCP2-AMPK-ACC signaling pathway of liver.
