Objective: To verify the content of S100A16 protein in human liver cancer tissues and hepatocellular carcinoma cells,and to preliminarily explore its role in hepatocellular carcinoma cell invasion and molecular mechanism,so as to provide a new molecular target for the treatment of liver cancer.Methods: GEPIA(http://gepia.cancer-pku.cn/)was used to query the expression of S100A16 protein in hepatocellular carcinoma and to query its survival analysis;Immunohistochemistry was used to detect the expression of S100A16 protein in hepatocellular carcinoma and paraneoplastic tissues;Using protein blotting to detect the expression of S100A16 protein in human hepatocellular carcinoma cell lines Hep-3b,Huh7 cells and normal hepatocytes HL-7702 cells;Construct S100A16 protein overexpression stable cell model and S100A16 protein knockdown cell model,using protein blotting and q RT-PCR to detect S100A16 protein and m RNA expression,and verify the successful infection;Use CCK8 assay,cell scratch assay,and Transwell invasion assay to investigate the effects of overexpression and knockdown of S100A16 protein on the proliferation,migration and invasion ability of hepatocellular carcinoma cell line Huh7 cells,respectively;Using GEPIA(http://gepia.cancerpku.cn/)to query the correlation between S100A16 protein and AKT,and Western blot to detect the expression of T-AKT,P-AKT expression,to initially explore the molecular mechanism of S100A16 protein in the proliferation,migration and invasion of hepatocellular carcinoma cells.Results: S100A16 protein was shown to be highly expressed in hepatocellular carcinoma tissues relative to paraneoplastic tissues on GEPIA,and the immunohistochemical results were consistent with this;Overexpression of S100A16 promoted hepatocellular carcinoma cell migration,proliferation and invasion.Knockdown of S100A16 protein was the opposite;Overexpression of S100A16 protein significantly elevated AKT phosphorylation,and knockdown of S100A16 protein did the opposite.Conclusion: S100A16 protein was highly expressed in liver cancer tissues and cells,and S100A16 protein promoted the proliferation,migration and invasion of liver cancer cells;S100A16 protein may promote the migration,proliferation and invasion of hepatocellular carcinoma cells by promoting AKT phosphorylation.
