ObjectivePrimary liver cancer are highly heterogeneous malignancy,with onset insidiousness and rapid progression.Hepatocellular carcinoma(HCC)is the most common neoplasm.Multiple microarray data in public database and Quantitative Real time PCR(qRT-PCR)were used to analyze the expression of C7 gene in Hepatocellular carcinoma and explore the correlation between the expression of C7 gene and the pathogenesis of HCC,clinicopathological parameters and prognosis.MethodsC7 gene,which has a significant expression difference in HCC,was obtained from the mRNA microarray datasets in GEO database.Hepatocellular carcinoma tissues and adjacent tissues were collected from 14 patients with HCC.qRT-PCR was used to detect the expression level of C7 gene.Referring to the clinical information from the TCGA database,the association between the expression level of C7 gene and the prognosis and clinical pathological parameters was analyzed.The bioinformatics analysis of C7 co-expressed genes which correlate with HCC were analyzed by applying the Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG).ResultsCompared to the noncancerous tissues,Meta-analysis showed the lower expression of C7 gene in tumor tissues.The data from TCGA indicated that C7 gene was down-regulation in patients with vascular invasion.It showed that patients with C7 high-expression in tumor tissues had significantly prolonged the survival time of OS and DFS,compared with those with low-expression.616 genes were as the co-expression genes of C7 gene for bioinformatics analysis and they were highly enriched in Focal adhesion,Focal adhesion pathway,PI3K/AKT signaling pathway and Pathways in cancer.ConclusionIt was proved that the expression of C7 gene was lower in HCC tissues than that in noncancerous tissues.The down-regulation of the C7 gene could predict adverse outcomes in patients with HCC and may offer a new therapeutical target for the treatment of HCC.New ideas for exploration of the pathogenesis of HCC may be provided by Bioinformatics analysis.
