Traditional Chinese medicine is a treasure of the Chinese nation that has been passed down for thousands of years.It has attracted more and more attention around the world because of its remarkable therapeutic effect.An herbal extract usually contains hundreds or thousands of chemical components,and these compounds may have significantly different polarities,quantities,and structures.Therefore,the comprehensive separation and characterization of the chemical components of Chinese herbal medicines has always been a major challenge in the field of separation science.High performance liquid chromatography(HPLC)has been proven to be the most commonly used and most effective method for separating complex chemical components of traditional Chinese medicine.However,traditional high performance liquid chromatography can only separate and detect multiple main compounds from one plant extract in one analysis.In recent years,the rapid development of two-dimensional liquid chromatography has provided new ideas for the separation of complex components in traditional Chinese medicine.Liquid chromatography can be divided into analytical chromatography and preparative chromatography according to the purpose of use.Analytical chromatography pays more attention to the separation effect,while preparative chromatography focuses on the separation and purification of sample components.In this paper,a set of online comprehensive two-dimensional reversed-phase chromatography×hydrophilic chromatography-mass spectrometry system constructed previously with multiple auxiliary modulations is used in the separation and analysis of traditional Chinese medicine ginseng and panax notoginseng.In the subsequent work,we will also successfully enlarge it.Up to the preparation level,the two-dimensional liquid phase preparation and separation of saponins in the traditional Chinese medicine ginseng and notoginseng has been realized.The specific research is as follows:1.Taking the traditional Chinese medicine Panax notoginseng and ginseng as the research objects,the online comprehensive two-dimensional reversed-phase chromatography × hydrophilic chromatography system was used to analyze the saponins in the two traditional Chinese medicines.Under exactly the same separation conditions,a full two-dimensional liquid phase analysis of the traditional Chinese medicine Panax notoginseng and ginseng extract was realized.After that,the twodimensional system is used in conjunction with mass spectrometry to detect the structure information of the compound,and the obtained mass spectrometry data is compared with existing references to confirm the obtained compound information.Afterwards,the difference of the saponins separated from Panax notoginseng was compared by mass spectrometry.2.In order to further study the application of the online comprehensive two-dimensional reversed-phase chromatography ×hydrophilic chromatography system in the separation of traditional Chinese medicine,we have scaled up the entire separation process to the preparative level and constructed a set of online comprehensive twodimensional preparative liquid chromatography system.Taking the ginseng extract as the sample,the preparation and separation of the saponin components in the ginseng extract were achieved by optimizing the separation gradient.Afterwards,in order to further improve the preparation efficiency,macroporous resin was used to remove impurities and concentrate the ginseng extract,and the sample was roughly separated to obtain ginsengtriol and ginsengdiol components.Due to the complex diol components,they were further used for the preparation of two-dimensional analysis,which was further optimized.After the diol preparation and separation conditions,28 ginseng diol saponin monomer fractions were finally obtained in a single preparation and separation process,and the structure was identified by mass spectrometry.The obtained mass spectrometry data was compared with the existing references to confirm the obtained compounds.information.3.The saponins contained in Panax notoginseng are similar to those in ginseng.Based on previous experiments,we used Panax notoginseng extract as a sample,optimized the separation gradient under the preparation conditions,and successfully prepared the full two-dimensional liquid phase.The chromatographic system is used for the preparation and separation of the traditional Chinese medicine Panax notoginseng.Under optimized conditions,30 panax notoginseng saponins monomers were obtained in one preparation and separation.The monomer structure was identified by mass spectrometry,and the obtained compound information was confirmed by comparison with existing references.Through the online comprehensive two-dimensional analysis and comprehensive two-dimensional preparation and separation of the traditional Chinese medicine Panax notoginseng and Panax notoginseng,the differences in the components of the traditional Chinese medicine Panax notoginseng and Panax notoginseng are preliminarily compared,and it also shows that the online two-dimensional analysis and preparative liquid chromatography established in this article The system has great potential for development in the analysis and separation of traditional Chinese medicine.
