| Objective: The rats with heart failure were established by intraperitoneal injection of adriamycin.The intervention of Yudanrongxin pill was conducted to study the effects of Yudanrongxin pill on left ventricular ejection fraction,serum natriuretic peptide,hypersensitive C-reactive protein and tumor necrosis factor-α in heart failure rats,and to explore the effects of Yudanrongxin pill on heart function and inflammatory factors in heart failure rats.Methods: Forty-five adult male SD rats(body weight 200±25 g)were fed in the laboratory for one week in advance.Thirty rats were randomly selected to form a model group,and the remaining 15 rats were used as the control group.The rats were divided into cages and fed.Model group: The rats were intraperitoneally injected with doxorubicin(2.5 mg/kg/W)every Tuesday and Friday,and 6 weeks were spent for modeling.The total amount of drug in each rat reached 15 mg/kg.The control group was injected intraperitoneally with the same volume of saline.One week after modeling,10% chloral hydrate(0.4 m L /100 g)was injected intraperitoneally to make the animals analgesic.LVEF of rats in each group was examined by cardiac ultrasound,and blood samples were collected from the left jugular vein of rats.ELISA was used to determine serum BNP level.All the rats survived during the modeling process.The rats were randomly divided into two groups: experimental group(N=15)and model group(N=15).Rats in the experimental group were given Yudan Rongxin pills(2.43 g/m L,2.43 g/kg.d)by gavage,while those in the control group and model group were given equal volume of normal saline by gavage.Four weeks after intervention,1 day after the last injection of drugs,the rats were given cardiac ultrasound examination under the same anesthesia,blood was then collected from the rats abdominal aorta.Serum BNP,hs-CRP and TNF-α levels of rats were determined by ELISA method.The pathological changes of myocardial HE staining were observed under microscope.The data of each group were recorded and statistically analyzed.Results:1.Compared with the control group,BNP obviously increases and LVEF significantly decreased in the rats with adriamycin-induced heart failure,with P<0.05,and the differences were statistically significant;2.After the completion of the test: compared with the control group,experimental group rats the BNP is still on the high side,P<0.05,the difference was statistically significant;Compared with model group,serum BNP in experimental group was decreased,P<0.05,the difference was statistically significant;3.After the completion of the test: compared with the control group,experimental group rats obviously low LVEF,P<0.05,the difference was statistically significant;Compared with model group,LVEF of the experimental group was increased,P<0.05,the difference was statistically significant;4.After the completion of the test: Compared with model group,the experimental group rats serum hs-CRP,TNF alpha decreased obviously,P<0.05,the difference was statistically significant;5.The model group complied with the cardiomyopathy sample changes,and it was significantly reduced in the experimental group rats.Conclusion: Yu Dan rong xin pill can reduce the serum levels of hs-CRP and TNF-α in rats with adriamycin-induced heart failure,decrease the myocardial inflammation and the pathological damage of myocardial cells,reduce the level of BNP,improve the cardiac function in heart failure rats,and have a protective effect on the heart. |
