The Effects Of Estrogen On Adriamycin-induced Heart Failure In Rats.

Objective: Now we have pay close attention to the protection effects of estrogen on the cardiovascular system. The concrete mechanism is still in studying. This experiment observe the protective effect of Estrogen on adriamycin-induced heart failure in rats.Methods: Thirty adult male Wistar rats were divided into 3 groups: A group, B group and C group. The B group and C group were intraperitoneal(IP) injection of ADR at a dose of 4mg/kg once a week for 6 weeks, the A group received an equivalent volume of sterile water. Estrogen(Diethylstilbestrol) 0.25mg/kg was administered using a stomach tube in C group every three days for 6 weeks. One week after completion of the ADR and Estrogen administration, the following parameters were measured using Buxco system: heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),±dp/dtmax. At the same time, body weights(BW),ventricular weight(VW) were evaluated. The Rats were killed after all measurements and the hearts were removed and processed for histological examination.Results: Compared with A group, the LVSP,±dp/dtmax of B Group decreased significantly(P<0.05), the LVEDP increase significantly (P<0.05);but the LVSP,±dp/dtmax of C group increased significantly(P<0.05) ,and the LVEDP decreased (P<0.05)compared with those of B group. Pathological examination demonstrated that the myocardial tissue of B group was damaged and the histological changes were consistent with those of cardiomyopathy. Heart pathological changes of the C group were wild.Conclusions: Estrogen can relieve the toxic effect of ADR on myocardium and greatly improve the contractivity of heart-failure in rats.
