Objectives:To explore the role of micro RNA-138(miR-138)in migration and invasion of gastric cancer(GC)cells and potential underlying mechanism.Methods:(1)Mir-138 mimics and Mir-138 inhibitor were transfected into HGC27 cells to detect migration and invasion in gastric cancer cells.(2)The regulatory effect of miR-138 on RhoC was verified by qRT-PCR and westen blotting.(3)RhoC gene expression in HGC27 cell line was silenced by si RNA,and the gene knockout was verified by RT-q PCR and Western blotting experiments.The effect of RhoC silencing on invasion and metastasis of gastric cancer cells was detected by scratch test and invasion test.(4)We investigated the effect of miR-138 on the protein expression of p-FAK,p-MAPK and p-ERK1/2 through Western Blotting;And the effect of RhoC silencing on the signaling pathway.(5)Cell migration and invasion experiments were used to verify that Mir-138 indeed regulates the migration and invasion of gastric cancer cells through RhoC/FAK/MAPK signaling pathway.Ressults:(1)MiR-138 overexpression inhibited HGC27 cells invasion and migration(P<0.05),while miR-138 knockdown has the opposite effect(P<0.05).(2)miR-138 negatively regulated the Ras homolog gene family,member C(RhoC)protein expression(P<0.05).(3)RhoC knockdown inhibited invasion and migration in HGC27 cells(P<0.05).(4)miR-138 overexpression attenuated FAK/MAPK pathway-related protein expression(P<0.05).(5)Additionally,RhoC silence and the inhibitors of FAK/MAPK pathway exert a partially reversible function in the miR-138 inhibition on invasion and migration in HGC27 cells(P<0.05).Conclusion:1.Mir-138 can inhibit the migration and invasion of gastric cancer cells.2.The inhibition of Mir-138 on the migration and invasion of gastric cancer cells was mediated by RhoC/FAK/MAPK signaling pathway.
