Malus hupehensis is a genus of apple trees belonging to the family Rosaceae.Its leaves contain flavonoids,tea polyphenols,proteins and various minerals and other chemical constituents.It has a variety of pharmacological activities,among which the flavonoids are the main activities of the leaves of Malus hupehensis.Ding Qiong and others isolated three hydroxyphloridzin,quercetin,and phloridzin from the total flavonoids of the leaves of Malus hupehensis which belong to dihydrochalcone with hypoglycemic,lowering blood pressure,Antioxidant,antibacterial,anti-tumor,osteoporosis prevention and anti-aging and many other biological activity functions.Traditionally,the leaves of Malus hupehensis is a regular drinking herbal tea and medicine.It is a dual-use medicinal herb and is contained in?Quality Standards for Hubei Chinese Medicinal Materials?(2009 edition).It has been reviewed and approved by the National Health and Family Planning Commission as a new food.At present,there is no comprehensive and systematic study on the characteristics of the effective components of the growth cycle of the leaves of Malus hupehensis;traditional methods of leaf treatment reduce the content of the active ingredients;there is currently no report on the application and literature of the preparation of the leaves of Malus hupehensis.It still stays at the stage of mainly processed products.In this way,the active ingredients are not easily dissolved out,the utilization rate is low and high waste.The purpose of this paper is to use supermolecular technology to develop a new type preparation product of the leaves of Malus hupehensis.This research includes that observe the growth characteristics of the leaves of Malus hupehensis in order to select the right time for picking herbs;optimize the collection and treatment techniques of leaf herbs;test the nature of the main components and explore the analytical methods;determine the analysis and preparation techniques of the active ingredient-cyclodextrin;optimize the prescription and the development of the product.To obtain high-quality medicinal materials,the HPLC was used to monitor the content of phloside and total polyphenols in the effective components of the leaves of Malus hupehensis,during different growth periods,the increase and decrease of biomass during the leaf growth period were comprehensively examined,the biomass of the active ingredients was determined and analyzed in order to choose the timing of picking the leaves;test the traditional baking and microwave drying treatment methods,compare the difference between the two active ingredients,analyse and compare the difference in vitro antioxidant properties,determine the best treatment,prepare for the preparation of high quality medicine raw material.Test the active ingredient properties,measure water-soluble phloridzin in the leaves of Malus hupehensis by UV-Vis spectrophotometry and method was validated,the possibility of the formation of supramolecule of active ingredients of the leaves of Malus hupehensis and cyclodextrin can be assessed by UV spectrophotometry and determine the binding constants,screen the supramolecular preparation(rotary and wet preparation,grinding speed,time and material ratio combinations)by orthogonal test,water-soluble phloridzin elution and the leaves of Malus hupehensis powder ratio as indicators to judge the best ratio,use scanning electron microscopy(SEM),UV-vis DRS,infrared diffuse reflectance(DR-FTIR),thermal analysis(DTA)and other technologies to verify the formation of ultrafine powder.Study the solubilization times and stability of ultrafine powders.Finally,the single-factor experiment was used to screen the prescription and technology of the leaves of Malus hupehensis micropowder tablets.The tablets were subjected to in vitro dissolution experiments and their stability was determined by influencing factors.The results of this paper found that the highest period of effective components of the leaves of Malus hupehensis was at the beginning of September.The content of active ingredients of the leaves picked by microwave drying was significantly higher than that of traditional treatments.Ultraviolet spectroscopy experiments show that the active components of the leaves of Malus hupehensis and cyclodextrin are easy to form supramolecular bodies;both dry and wet methods can improve the dissolution of phlorizin,but the dry method is better;the mass ratio of CD is 1.5:1,and the dry grinding conditions is the mass ratio of the grinding material is 20:1,the rotating speed is 450 r/min,and the time is 12 min.The powder suitable for preparing the preparation can be obtained under this condition;DTA,UV-vis DRS,DR-FTIR,and TEM showed that a new structural system was formed between the active components of the leaves of Malus hupehensis and ?-CD.The ultra-fine powder of the leaves of Malus hupehensis containing ?-CD inclusion complex was prepared.The amount of phloridzin eluted by the brewing method increased by more than 3 times and the photothermal stability was significantly improved.The formulation of the tablets was 84%of the leaves ultrafine powder,10%of microcrystalline cellulose,and 5%of the state of the art.Bubbling,75%aqueous ethanol solution,1%magnesium stearate,tablet pressure 4-6 Kg,total tablet weight 0.65 g;Influencing factor testing showed that the self-made the leaves of Malus hupehensis powder tablet has good stability under the test conditions.powder of Malus hupehensis which provides new ideas and methods for the deIn summary,the combination of ultrafine crushing with inclusion technology developed the tablets of microvelopment and utilization of the leaves of Malus hupehensis resources.
