Benzo[a]pyrene(BaP),the most toxic polycyclic aromatic hydrocarbon,is a global food-borne pollutant,and is widely presented inedible oils,baked bread,barbecue and smoked foods,could lead to severe liver injury and intestinal microbial disorders and so on.Isoorientin(ISO),a natural flavonoid compound that is widely found in various foods,and possesses multiple biological activities including anti-inflammatory,anti-cancer,antioxidant,anti-diabetic,hepatoprotective effect and so on.However,the effect of ISO on liver injury and intestinal microflora disorders induced by BaP remains unknown.Therefore,ISO was seen as the study subject in this study.And the objective of this study is to investigate the effect of ISO on liver injury and intestinal microflora disorders of male BALB/c mice exposed to BaP.Our study aims to provide a theoretical basis for the prevention of BaP-induced toxicity and the potential value of ISO.Main results for this study are as follows:1.ISO could effectively improve the weight change of mice.Observing the performance and measuring the weight food intake,F/G,and liver index of mice indicated that ISO could relive the impact on growth performance of mice exposed to BaP by increasing the daily food intake and weight gain,reducing the F/G and liver index to promote the healthy growth of mice.2.ISO could markedly relieve the liver injury caused by BaP.The results of liver function index and oxidation index showed that ISO could alleviate BaP-induced liver dysfunction and oxidative damage by decreasing the levels of ALT and AST,enhancing the activity of T-SOD and GSH-Px,down-regulating the levels of MDA and H2O2.The HE staining results showed that ISO could inhibit the liver tissue injury induced by BaP through increasing the number of cells and narrowing the intercellular space.Transmission electron microscopy,RT-PCR assays and WesternBlot tests indicated that ISO could prevent BaP-induced autophagic and pyroptotic liver injury by down-regulating the numbers of autophagosomes,the mRNA expressions of LC3II and Beclin-1,LC3II,Beclin-1,IL-1? and IL-18,reducing the expression of Caspase-1 and decreasing the NO release.3.ISO could obviously alleviate and improve intestinal microflora disorders caused by BaP.16S rRNA gene sequencing results showed that ISO could improve the abundance of Platts and Lactobacillus,inhibite the increase in the abundance of Desulfurium,Acinetobacter,Bacillus and Veillonellain the gut flora,while inhibiting the increase in the metabolic pathways of lipopolysaccharide and sulfide to improve the intestinal microflora disorders caused by BaP.In addition,this chapter found that ISO could be able to prevent colonic oxidative injury caused by BaP by increasing the activity of T-SOD and GSH-Px in the colon and decreasing the levels of MDA and H2O2.The HE staining results also indicated that ISO could protect the colon tissue by decreasing the cellularity in crypts.This subject systematically studied the effects of ISO on liver injury and intestinal microflora disorders induced by BaP in vivo,and provided a scientific basis for terminating the injury induced by BaP and fully developing ISO resources.
