In this dissertation,we used chromatographic method and various conventional separation and purification methods to search for the chemical constituents of plant against tobacco Mosaic virus in Nicotiana glutinosa L and Gardneria ovate wall.1 Studies on Chemical Constituents from Nicotiana glutinosa LIn order to search for natural products with bioactivities,the 23 compounds were isolated and identificated from Nicotiana glutinosa L by using chromatographic methods,and the silica gel column chromatography,sephadex LH-20,MCI,RP-18 column chromatography,semi-preparative RP-HPLC were used for routine purifications.These structures were determined by spectral methods,including NMR,MS,UV,IR analysis and comparison with data in the literatures.The names of these isolations were naphthalenecarboxylate A(1),2-(pyridin-3-yl)-6-(2-(pyridin-3-yl)pyridin-5-yl)piperidine(2),labdan-8?-ol-15-yl-(labdanolate)(3),stigmasterol(4),?-ecdysterone(5),capitasterone(6),?,?-dipyridyl(7),N,N-bis(2-chloroethyl)-N-methyl-N-(2-nitrobenzyl)ammonium chloride(8),[S-(2exo,3exo)]-1,7,7-Trimethyl-3-[methyl[(pyridin-3-yl)methyl]amino]bicyclo[2.2.1]heptan-2-ol(9),lycoranine D(10),isoscopoletin(11),ent-8a,9b-dihydroxylabda-13(16),14-dien-1-one(12),penienone(13),3-O-(2,3-dimethylbutanoyl)-13-O-dodecanoyl-20-deoxyingenol(14),ataloxime B(15),wenyujinin L(16),(+)-8,9-epoxyselina-4,11-diene(17),19-nor-abieta-4(18),8,11,13-tetraen-7-one(18),aloesaponarin II(19),aminobenzoates C(20),N'-Ethylnornicotine(21),nicotine(22),3-methylindan-1-one(23).Compounds(1-23)are reported for the first time from this plant.2 Studies on Chemical Constituents from Gardneria ovate wallThe genus Gardneria,belonging to the Family Loganiaceae,and Gardneria ovata Wall.is a climbing shrub distributed in Guangxi,Yunnan,and Xizang provinces of China.Previous phytochemical investigations revealed that the Gardneria plants are good sources of mono-and bis-terpenoid indole alkaloids with diverse skeletons.To the best of our knowledge,there are few investigations on the chemical constituents of G.ovate.A chemical investigation on G.ovate led to the isolation of14 compounds by using silica gel column chromatography,sephadex LH-20,MCI,RP-18 column chromatography,semi-preparative RP-HPLC and so on.These structures were determined by spectral methods,and named as:19(E)-9,12-didemethoxy-11-methoxy-16-dehydroxylchitosenine-17-O-?-D-glucopyra noside(24),19(E)-9-demethoxy-16-dehydroxylchitosenine-17-O-?-D-glucopyranoside(25),19(E)-9,18-didemethoxy-gardneramine(26),19(E)-11-methoxy-9,18-didemethoxygardneramine(27),19(E)-18-demethoxygardneramine(28),gardfloramine-9-O-?-D-glucopyranoside(29),gardfloramine(30),9-demethoxy-18-demethylgardneramine(31),gardneramine(32),18-demethylgardneramine(33),gardneramine iminoether(34),chitosenine(35),gardmutine D(36),gardmutine A(37).Compounds 24 is a new constituent.Compounds 24,25,34-37 were reported for the first time from Gardneria.3 The anti-TMV activity of the isolationsAll the isolations(37 compounds)were evaluated their anti-TMV activity.The results showed that the inhibition rate of compounds 15?12?13?17?24?25 and 26 were more than 60%,equal to positive control.compounds 3?4?6?8?9?11?28?29?31?34 were more than 55% also have potential effect against tobacco Mosaic virus(TMV).
