The identification of sugar beet fertility is of practical importance to select superior germplasm resources for breeding work.The use of marker-assisted selection(MAS)can significantly improve the efficiency of sugar beet fertility identification.This study was conducted to identify the fertility of sugar beet multigerm and monogerm germplasm resources.The study was to analyze the breeding composition of sugar beet multigerm germplasm resources by MAS.To optimize the whole identification process of sugar beet breeding using molecular markers.To analyze the breeding gene composition of sugar beet monogerm sterile lines and maintenance lines.The study was performed to identify sugar beet sterile cytoplasm(S)and normal cytoplasm(N)by the polymorphism of variable number of tandem repeats(VNTR)loci.The genetic composition of the fertility genes was analyzed by linking the molecular markers s17 and o7 to the sugar beet nucleus fertility recovery genes Rf1 and Rf2.At the same time,seven pairs of primers designed according to the difference between sugar beet genome and transcriptome were screened and validated.The results showed that the 627 sugar beet multigerm germplasm resources had complex breeding composition,with three cytoplasmic types and two Rf1 genotypes.Optimization of the process for identifying sugar beet breeding showed that the lysate method for extracting sugar beet DNA had greater advantages over the CTAB method and alkaline lysate;the 6% non-denaturing polyacrylamide gel electrophoresis was not suitable as the fertility identification.the use of dual PCR for sugar beet fertility identification was completely feasible,while triple PCR was not.The molecular marker s17 associated with the Rf1 locus and the enzyme digestion verification can be useful for the identification of sugar beet maintainers and sterile lines,while the Rf2 locus may play a minor role in the recovery of sugar beet fertility.In addition,primer 11E8M4 S was suitable for the identification of sugar beet maintainers and sterile lines.Through this study,we have a preliminary understanding of the breeding composition of multigerm sugar beet in the sugar beet germplasm repository,and the applicability of different molecular markers is elucidated.Optimizing the whole identification system for the identification of sugar beet breeding by molecular markers,which greatly reduces the identification time and cost and lay the foundation for sugar beet variety improvement in the future.
