Research On The Combination Of Baibu, Cnidium, Phellodendri And Other Special Plant Compounds For Flue-cured Tobacco Planting

The Botanical Mixture was mainly composed of Stemona japonica,Cnidium monnieri(L.)Cuss.,Cortex Phellodendri,and other herbs,dedicatedly used in tobacco planting.When it was applied in filed,water should be used as solvent,and brewed by gentle heat,it has antiviral,antibacterial,and growth-promoting effect.In this study,UV,HPLC and GC-MS was adopted to analyze chemical composition of the water decoction of Botanical Mixture,The main results were as follows:1.The qualitative identification of the experiental showed the water decoction of Botanical Mixture contains alkaloids,flavonoids,coumarins,organic acids and other steroidal saponins.The content of total alkaloids,flavonoids and coumarins in water decoction of Botanical Mixture were determined by UV,16.5 g Botanical Mixture contained 62.82 mg alkaloids,409.30 mg flavonoids and 176.62 mg coumarins.The distribution of these three substances in different polar part were Comparative analyzed.The distribution of total alkaloids were as follows: Ethyl acetate phase was8.47 mg,n-butanol phase was 31.07 mg,water phase was 13.66 mg.The distribution of total flavonoids were as follows: Ethyl acetate phase was 87.52 mg,n-butanol phase was 150.67 mg,water phase was 157.31 mg.The distribution of coumarins were as follows: Ethyl acetate phase was 42.63 mg,n-butanol phase was 76.25 mg,water phase was 31.90 mg.2.Alkaloids,coumarins and other chemical components in the water decoction of Botanical Mixture were analyzed by HPLC.Alkaloids such as matrine,Oxymatrine,sophoridine,Jatrorrhizine hydrochloride,Palmatine chloride and berberine hydrochloride were found in the water decoction of Botanical Mixture.Coumarins contains imperatorinand osthole were found in the water decoction of Botanical Mixture,as well as Chlorogenic acid.3.The content of three major components of matrine,Oxymatrine and berberine hydrochloride in Botanical Mixture were determined by HPLC.The results indicated that 16.5 g Botanical Mixture herbs was extracted by water boiling which content of matrine,Oxymatrine and berberine hydrochloride contents were 7.67 mg,9.78 mg,9.78 mg respectively,In total 23.59 mg4.The volatile components of the water decoction of Botanical Mixture,volatile oil of Botanical Mixture and the solution after volatile oil extraction of Botanical Mixture were determined by GC-MS.The detection results of the water decoction of Botanical Mixture,volatile oil of Botanical Mixture and the solution after volatile oil extraction of Botanical Mixture witch their contents of volatile components were71,119 and 121 respectively,the chemical composition be retrieved were 34,69 and 60 respectively.The solution after volatile oil and the water decoction of Botanical Mixture extraction of Botanical Mixture included three common compositions Pentadecane,Dihydroactinidiolide and 3-methyl-7-methoxy-2-benzopyran-1(1H)-one;compared with volatile oil of Botanical Mixture,twenty-one compositions them possessed,such as ?-Pinene,Camphene,Artemisia alcohol,Camphor et al.But both of volatile oil of Botanical Mixture and water decoction of Botanical Mixture had no same composition in retrieved.
