ObjectiveThe correlation between acupoints and zang-fu organs is one of the core contents in the acupuncture basis theory since the "75"plan was put forward.Previous studies focused on the regulation of meridian acupuncture on the functions of the corresponding viscera.Acupuncture and moxibustion has two-way adjustment effect on circulatory system,endocrine system,digestive system and respiratory system.There are also some researches focus on the reflect of body surface acupoints when the organs are in pathological injury state,pathological changes often occur in body surface acupoints,such as abnormal shape,temperature change and resistance imbalance.However,the changes of related body surface acupoints after acupuncture for regulating viscera are rarely reported.Based on the animal model of myocardial ischemia,it is found that low frequency electroacupuncture can improve the disturbance of cardiac hemodynamics after myocardial ischemia,promote the normal metabolism of myocardium and adjust the contractile force of myocardium through the study of different levels of cardiac electrophysiology,histomorphology,hemodynamics,myocardial enzymes and cardiovascular active substances to recovery of myocardial function.The research focuses on the regulation effect and mechanism of low frequency electroacupuncture on target organs.After the intervention of low frequency electroacupuncture,there is no report on the reflection of body surface meridians.In this study,Changes of microcirculation blood perfusion in pericardial meridians were detected by LSCI after low frequency electroacupuncture at acupoints or non acupoints with rats in myocardial ischemia.According to the results of blood perfusion of acupoint microcirculation,the tissue of the point area was selected to detect the content of nitric oxide and calmodulin..From the perspective of sympathetic nerve mediated nitric oxide and calmodulin changes,the mechanism of the difference of blood flow volume between different points of pericardial meridians was discussed.Methods60 male Wistar rats were randomly divided into the one times group(lt group)and three times group(3t group),and each group was divided into control group,sham-operation group(a line went through the left anterior descending branch of coronaryartery,non-ligation),the model group(ligation of the left anterior descending branch of coronary artery),the low-frequency EA group and the low-frequency EN group.Electroacupuncture 1 times a day,each time 20min,frequency of 2Hz.After the first or third time of EA treatment,the ECG was detected by BL-420E+,and skin blood perfusion was tested by LSCI around "Neiguan" and"Ximen "and" Tianquan".Data was analyzed The skin tissue of PC 6,PC 4,PC 2 were taken on ice,Based on the result of infrared thermal imaging,selected the specific acupoint on temperature change,The content of NO metabolites nitrate NO3-and NO2-by Nitrate reductive enzymatic to indirectly reflect NO contents.The content of CaM were detected by ELISA.Results(1)Comparison between the groups after 1 times treatmentCompared with the control group,the J-point amplitudes of model groups increased significantly(P<0.01).Compared with the model group,the J-point amplitudes of the low-frequency EA group were significantly reduced(P<0.01),the J-point amplitudes of the low-frequency EN group had no significant difference.the low-frequency EA group and the low-frequency EN groupCompared with the normal group,the skin blood flow changes of"Neiguan" and"Ximen" and "Tianquan" were significantly decreased in model group(P<0.01).Compared with the model group,the skin blood flow changes of "Neiguan" and "Ximen" were significantly increased after 1 times EA treatment(P<0.01),but after 1 times EN treatment there were no obvious change.the skin blood flow changes of "Tianquan"was no obvious change after 1 times EA treatment and EN treatment(P>0.05)..Compared with the normal group,the NO and CaM content of "Neiguan" and"Tianquan" were significantly decreased in model group(P<0.01).Compared with the model group,the NO and CaM content of "Neiguan" were significantly increased after 1 times EA treatment(P<0.01),but there were no obvious change after 1 times EA treatment.the NO and CaM content of "Tianquan" were no obvious change after 1 times EA treatment and EN treatment(P>0.05).(2)Comparison between the groups after 3 times treatment:Compared with the control group,the J-point amplitudes of model groups increased significantly(P<0.01).Compared with the model group,the low-frequency electroacupuncture point group were significantly reduced(P<0.01).There was no significant difference between low-frequency electroacupuncture group and high-frequency electroacupuncture group(P>0.05).The law of change between three and one times group is consistent.Compared with the normal group,the skin blood flow changes of "Neiguan" and"Ximen" and "Tianquan" were significantly decreased in model group(P<0.01).Compared with the model group,the skin blood flow changes of "Neiguan" and "Ximen" and"Tianquan" were significantly increased after 3 times EA treatment(P<0.01),but there were no obvious change after 3 times EN treatment.Compared with the normal group,the NO and CaM content of "Neiguan" and"Tianquan" were significantly decreased in model group(P<0.01).Compared with the model group,the NO and CaM content of"Neiguan" and "Tianquan"were significantly increased after 3 times EA treatment(P<0.01),but there were no obvious change after 3 times EN treatment.(3)Comparison between the It group and the 3t groupCompared with1t group,"Tianquan" skin blood flow and area of NO and CaM the low-frequency EA group were significantly increased in the low-frequency EA group(P<0.01),while there was no obvious change in the low-frequency EN groupConclusion(1)The rats in myocardial ischemia state,acupoints skin blood flow decreased obviously and there is a significant difference in the degree of change of the pericardium through different acupoints.The rats in low-frequency EA state,acupoints skin blood flow of pericardium increased significantly,which the change of "Tianquan"blood flow was effected by the number of treatment.(2)The low frequency electroacupuncture had a certain repair effect on the myocardial ischemia inj ury.(3)The changes of NO and CaM contents in the skin tissue of the pericardial meridian area were consistent with the change of acupoint blood flow.It indicates that after the regulation of the myocardium by low frequency electroacupuncture,the blood flow characteristics on the surface of the body surface appear,which is related to the changes of NO and CaM in the sympathetic nerves.
