Rhizoma Corydalis is a traditional Chinese medicinal material and is the dried tuber of Corydalis yanhusuo W.T.Wang,a perennial herb belonging to family Papaveraceae.In recent years,more and more research on Rhizoma Corydalis have been published and the market demand has been increasing.Rhizoma Corydalis is mainly used in the treatment of dull pain such as abdominal pain,amenorrhea,postpartum stasis,and swelling pain,but its mechanism is still unclear.At present,there are some misunderstandings and confusion in the market about its quality grades.The wild resources of Rhizoma Corydalis are scarce and the material sold in the market mainly comes from artificial cultivation.The analysis of the differences from different producing areas and the causes of the differences will provide the basis for the production of high quality Rhizoma Corydalis.Based on this,this paper discusses a rapid and accurate method for the simultaneous determination of 8 alkaloids in Rhizoma Corydalis was established,which was used to detecte the contents of 8 alkaloids in Rhizoma Corydalis with different sizes and specifications.The detection results provided theoretical basis for the classification of commodity grades of Rhizoma Corydalis in the market.The differences of 8 alkaloid contents in periderm,phloem and xylem of Rhizoma Corydalis were detected and analyzed.Rhizoma Corydalis cultivated in Zhejiang and Shaanxi provinces were collected and sorted out.The geographic detector was used to study the influence degree and difference of different climatic factors on the content of 8 alkaloids in Rhizoma Corydalis.The mechanism of Rhizoma Corydalis in the anagesia pain based on network pharmacology.The main research contents and results are as follows:1.A fast and accurate high-performance liquid phase method was developed for the simultaneous detection of 8 alkaloids in Rhizoma Corydalis.The content of eight alkaloids in five different sizes of Rhizoma Corydalis was detected.It was found that the content of 7 alkaloids decreased with the increase of the diameter of Rhizoma Corydalis,while the content of protoopioids increased with the increase of the diameter.The total content of 8 alkaloids decreased with the increase of the diameter of Rhizoma Corydalis.2.The tuber of Rhizoma Corydalis was separated into periderm,phloem and xylem three parts.The content of 8 alkaloids in these three parts was measured.It was found that there were significant differences in the content of eight alkaloids in the three parts,among which the content of alkaloids in periderm was the highest,that in phloem was the middle,and that in xylem was the lowest.The measurement results explain the reason why the total alkaloid in Rhizoma Corydalis increases as the diameter of Rhizoma Corydalis decreases.3.The alkaloid content in tubers,stem and leaf of C.yanhusuo during growth periods was determined.It was found that the alkaloid content in Rhizoma Corydalis in different growth periods was different and the content of alkaloids in the mother tuber has a significant decrease during the period of plant development.The tetrahydropalmatine and glaucine were contained in the stems and leaves,and the content of glaucine in the stems and leaves was higher than that in the tubers.The results provide information for the development and utilization of the aerial parts of C.yanhusuo.4.The samples of Rhizoma Corydalis from 13 regions in Zhejiang Province and Shaanxi Province and the related climatic and environmental factors were collected.The content of 8 alkaloids in Rhizoma Corydalis from 13 regions was measured.To what extent the climate factors influence spatial differentiation of the content of 8 alkaloids in Rhizoma Corydalis was explained by using geographical detector.This paper analyzed the interaction detection and risk detection among the monthly average temperature,monthly average precipitation,and monthly average relative humidity,which have the greatest influence on alkaloid accumulation.Then the climatic level suitable for alkaloid accumulation is selected and it was found that the maximum accumulation of four alkaloids of protopine,dehydrocorydaline,glaucine,tetrahydropalmatine,and total alkaloids was obtained under the condition of monthly average temperature 0.8-6?,monthly average relative humidity 60%-65%,and monthly average precipitation 4.8-50 mm.5.Based on network pharmacology,the mechanism of Rhizoma Corydalis in anagesia was discussed.The possible mechanism of 10 active ingredients such as berberine,scoulerine,D-tetrahydroj atrorrhizine,corybulbine,D-glaucine,tetrahydropalmatine,corydaline,tetrahydroberberine,isoboldine,and palmatine in Rhizoma Corydalis on traditional analgesic effect were explained preliminarily.17 targets related to the anagesia were screened and the "chemical component-target-pathway-disease" network was constructed.The results indicated that 10 active ingredients in Rhizoma Corydalis may play a role through multiple targets and different pathways.It provides a reference for further research on the analgesia mechanism of Rhizoma Corydalis in the future.The results of this paper provide theoretical basis for not only the reasonable grading of Rhizoma Corydalis sold in the market,but also the selection of planting area.Besides,this paper laid a foundation for increasing planting area and improving the yield of Rhizoma Corydalis.
