Objective:By observing the changes in m RNA expression of Rho A/ROCK and adenosine triphosphate binding cassette transporter A1(ABCA1)in the abdominal aorta of AS rabbits,and the content of lecithin cholesterol acyltransferase(LCAT)in serumto.To study the mechanism of treating atherosclerotic vulnerable plaques with herb medicine cake moxibustion.Methods:Seventy male New Zealand white rabbits were randomly divided into two groups after one week of adaptive feeding.One group is K blank group(blank control group)with 10 rabbits,the other group is surgery group with 60 rabbits,the operation group received balloon injury of abdominal aorta.The rabbits survived successfully and randomly divided into 5groups: M model group(AS vulnerable plaque model group);Z direct moxibustion group(moxa cone direct moxibustion on + model group);G herb medicine cake moxibustion group(herb medicine cake moxibustion+ model group);A drug group(Atorvastatin + model group);Y inhibitor group(Fasudil Hydrochloride+ model group).The blank group was fed with normal diet,and the M group was fed with high-fat diet,the two groups were not operated,only bundled.Z,G,A and Y groups began to feed with high-fat diet,and took corresponding treatment measures such as direct moxibustion,herb medicine cake moxibustion,oral atorvastatin,ear vein injection of fasudil,etc.for 8 weeks.After 8 weeks of treatment and intervention,local gene transfection was performed in the abdominal aortic plaque through the femoral artery,injecte Recombinant Human P53 Adv Injection(Ad5-p53,also known as Gendicine)into the largest plaques to establish the vulnerable plaque model of AS rabbits.The rabbits in M,Z,G,Y and A group are treated with drug-induced plaque rupture twice at24 and 48 hours before killed:Subperitoneal injection of Chinese Russell’s Viper Venom(CRVV)0.15 mg/kg and auricular vein intravenous injection of Histamine 0.02mg/kg.The contents of TC,TG,LDL-C,HDL-C,Apo-A,Apo-B and LCAT in blood were determined by enzyme linked immunosorbent assay ELISA.The m RNA expressions of Rho A,ROCK1 and ABCA1 in atherosclerotic vulnerable plaques of abdominal aorta in AS rabbits were detected by fluorescence quantitative PCR.Results:1.The level of blood lipid in rabbits: Compared with blank group,the contents of TG,TC,LDL,Apo-B and the ratio of Apo-B/Apo-A in model group were increased,while the content of HDL,the ratio of HDL/LDL and Apo-A were decreased(P < 0.01);Compared with model group,the contents of TG,TC,LDL,Apo-B and the ratio of Apo-B/Apo-A in direct moxibustion group,herb medicine cake moxibustion group and drug group were decreased,while the content of HDL,the ratio of HDL/LDL and the content of Apo-A were increased(P < 0.05 or P < 0.01);Compared with the direct moxibustion group,the content of APO-B in the herb medicine cake moxibustion group was significantly decreased(P < 0.01).The results showed that direct moxibustion,herb medicine cake moxibustion and the use of medicine atorvastatin were effective in the control of blood lipid in AS rabbits.The herb medicine cake moxibustion had the best effect in regulating the content of Apo-B compared with other treatment groups,and there is no difference between the three groups in regulating the content of TG,TC,LDL,HDL and Apo-A(P > 0.05).2.Expression of Rho A,rock1 and ABCA1 m RNA in abdominal aorta: Compared with the blank group,the expression of Rho A and ROCK1 m RNA in the model group increased,and the expression of ABCA1 m RNA decreased(P<0.05 or P<0.01);Compared with the model group,the expression of Rho A and ROCK1 m RNA in each treatment group decreased(P<0.05 or P<0.01);the expression of ABCA1 m RNA in each treatment group increased(P<0.05 or P<0.01).It shows that herb medicine cake moxibustion can inhibit the expression of Rho A and ROCK1 m RNA in VAP rabbits,and promote the expression of ABCA1 m RNA,Improve the level of reverse cholesterol transport and stabilize AS vulnerable plaques.3.Serum LCAT content: Compared with the blank group,the serum LCAT content of the model group was significantly reduced(P<0.05);compared with the model group,the serum LCAT content of each treatment group increased(P<0.01);Compared with the direct moxibustion group and the drug group,the content of LCAT in the herb medicine cake moxibustion group increased(P<0.05).It shows that herb medicine cake moxibustion can increase the LCAT content in AS rabbit serum.Conclusion:1.Herb medicine cake moxibustion has a certain regulatory effect on the blood lipids of AS rabbits2.Herb medicine cake moxibustion has a certain stabilizing effect on the vulnerable plaques of AS rabbits3.The mechanism of herb medicine cake moxibustion stabilizing vulnerable atherosclerotic plaque may be related to inhibiting Rho A /ROCK1 m RNA expression,up regulating ABCA1 m RNA expression,increasing LCAT content,increasing the level of reverse cholesterol transport is related.
