Objective:Observation of moxibustion with medicinal cake reverse transport effect on rabbit AS vulnerable plaque TLR2/4-IRF3signal transduction pathway in regulation of cholesterol, and expounds the mechanism of herbal cake separated moxibustion stability of vulnerable atherosclerotic plaques in protein level, for the future clinicalapplication of herbal cake separated moxibustion treatment and prevention of AS and provide experimental basis.Method:60New Zealand rabbits were, half male and half female, were randomly divided into5groups, namely:A blank group (control group); B model group (AS model group); C direct moxibustion group (AS model+direct moxa cone moxibustion on); D herbal cake separated moxibustion grou-p (AS model+herbal cake separated moxibustion the drug group); E (AS model+atorvastatin),12rats in each group. The establishment of AS vulnerable plaque of rabbit model by feeding high cholesterol and abdominal aorta balloon injury, methods, methods of treatment were given direct moxibustion, moxibustion, atorva-statin, continuous treatment for8weeks.Index and detection method:take the carotid artery blood, the determination of TC, TG contents after treatment with enzyme method determination of HDL-C, LDL-C colorimetry, the determination of apoA, apoB content ofelectrophoresis using transmission electron microscope; HE staining method to detect abdominal aorta intima and the thickness of lining in the film; gray usingimmunohistochemical detection of abdominal aorta LXR, ABCA1, TLR2, TLR4, IRF3values, observe the antigenimmune activity; using fluorescence quantitative PCR detec-tion technology, effects of herbal cake separated moxibustion on the expression of AS in rabbit vulnerable plaque TLR2, TLR4, IRF3mRNA.Results:1lipid content:compared with the model group, the serum TC, TG, LDL-C, Apo-B contents were significantly decreased (P<0.05or P<0.01), HDL-C, Apo-A content were increased significantly (P<0.05or P<0.01). Gray2abdominal aortic LXR, ABCA1, TLR2, TLR4, IRF3values:direct moxibustion group, moxibustion group and drug group were the Atherosclerotic Vulnerable Plaque of abdominal aorta in rabbits LXR, ABCA1gray value was significantly reduced (P<0.01), TL-R2, TLR4, IRF3gray value increased significantly (P<0.01or P<0.05) andisolation Herbal Cake separated moxibustion group, d-rug group is better than the direct moxibustion group.Expression of3.AS, TLR4, TLR2in vulnerable plaque in rabbit IR-F3mRNA:direct moxibustion group, moxibustiongroup and a medication group made AS vulnerable plaque in abdominal aorta of rabbits TLR2, TLR4, IRF3mRNAexpression was significantly down regulated (P<0.01or P<0.05), and herbal cake separated moxibustion gr-oup,drug group is better than the direct moxibustion group.4.AS vulnerable plaque intima-media thickness:compared with the blank group, model group, the thickness ofintima of rabbit aortic intima-media thickness, carotid intima media thickness (IMT) were significantly increased(P<0.01).Compared with the model group, moxibustion gr-oup, drug group and direct moxibustion group of rabbit aortic intima media thickness, carotid intima media thickness were significantly smaller(P<0.01), the increase in film thickne ss (P<0.01). Compared with the direct moxibustion group, moxibustion group and a medication group of rabbit aortic intima media thickness, carotid intima media thickness were significantly decreased (P<0.01or P<0.05). The cake separated moxibustion group compared with the medicine group, no significant difference between the two groups effect on endometrial thickness, intima-media thickness (P>0.05).5.AS rabbit abdominal aorta ultrastructure of vulnerable plaques:blank group endometrium in normal membrane structure; modelgroup of rabbit aortic wall destruction, endothelial and subendothelial severity and necrosis, intimal thickening, the surface has the rich foam cell coverage, a large number of lipid droplets and inflammatory cell infiltration, fibrous cap thinner, showing the characteristics of vulnerable plaque elastic fibers, the internal elastic membrane andmembrane rupture and collapse, smooth muscle cell infiltration; damage degree of direct moxibustion group aortic wall significantly reduced, endothelial cells than normal, but still found inside a small amount of foam cells and lipiddeposition, the internal elastic membrane and the membrane was complete; Every cake moxibustion medicine group, the drug group rabbit aortic endothelial cells complete, mostly normal form, the stretch film thickness uniformity in a row, mitochondria edema, occasional endothelial cells in the membrane. Conclusion:1.herbal cake separated moxibustion has a good regulating effect on AS vulnerable plaque of rabbit blood lipid and lipoprotein metabolism.2.herbal cake separated moxibustion can reduce the improve AS vulnerable plaque of rabbit aortic intimastructure, protection of aortic endothelial, inhibit intimal hyperplasia, reduce the IMT.3. Every cake moxibustion medicine can inhibit rabbit AS vulnerable plaques in TLR2and TLR4, IRF3antigen activity and TLR2and TLR4, IRF3mRNA expression and activity of LXR, ABCA1antigen, have the effect of stabilization AS vulnerable plaques.
