| Objective:Based on the difference in the expression of lncRNA H19 in the process of promoting the healing of diabetic ulcer wounds by the method of nourishing yin and regaining yang,and trying to analyze the difference between the method of nourishing yin and regaining yang in promoting the healing of diabetic ulcers.Methods:In the early stage of this project,the most representative lnc RNAs were selected through the selection of differentially expressed lnc RNAs screened by lnc RNA high-throughput sequencing technology and q-PCR verification;at the same time,animal experiments were carried out,and animal experiments were collected on the 5rd,7rd,and 10rd days,the granulation tissues of the acute group,mo del group,nourishing-yin group,and huiyang group were detected by real-time fluorescence quantitative PC R on the finally selected lnc RNA,and the detection results were analyzed.Results:1.After modeling,mice in the model group,nourishing-yin group,and huiyang group are obvious signs and symptoms of diabetes,because there are significantly more water intake,food intake,and urine output than those in the acute group.They are thinner,having wet hair,and having a heavier smell in the cage.Compared with the control group,the random blood glucose values on the 3rd,5rd,and 7rd days were significantly higher in the model group,nourishing-yin group,and huiyang group(p<0.01);compared with the model group.Nourishing-yin group and huiyang were no significant difference between the groups(p>0.05).2.From the perspective of eye observation,the wounds in the acute group were fresh and clean,with obvious blood vessels and fewer secretions.In the model group,a piece of yellow pus was formed,the wound surface was white,and the secretion was thicker.There were more secretions in the nourishing-yin group and the huiyang group,and the wounds were whitish.3.As time changes,the wound healing rate at each time point has a significant difference(●●p<0.01).On the 5rd,7rd,and 10rd days,the wound healing rate of the model group,nourishing-yin group,and huiyang group was significantly lower than that of the acute group(**p<0.01);while on the 5rd day,the model group,nourishing-yin group and Huiyang group were no significant difference in wound healing rate(▲p>0.05,■p>0.05);on the 7rd day,the wound healing rate of the nourishing-yin group and the Huiyang group was significantly higher than that of the model group(△△ p<0.01),the wound healing rate of the Huiyang group was significantly higher than that of the nourishing-yin group(□□p<0.01);on the 10rd day,the wound healing rate of the nourishing-yin group and the Huiyang group was higher than that of the model group(△ p<0.05)),there was no significant difference in wound healing rate between the Huiyang group and the nourishing-yin group(■p=0.869>0.05).4.ENST00000443 3 64,ENS T00000576797,ENST00000439725 q-PCR verification results are consistent with the chip results.But based on the amplification curve and melting curve ENS T00000439725 effect is better,and finally selecteing ENS T00000439725 for real-time fluorescent quantitative PC R detection.5.With the change of time,the expression of H19 in the acute group first decreased and then increased(p<0.01);the expression level of the model group had no significant difference(p>0.05);the expression level of the Huiyang group on the 5th day and the 7th day were no significant difference,and On the 10th day,that was lower than before(p<0.05);the expression of H19 in the nourishing-yin group continued to rise(p<0.01).On the 5rd day,the expression of H19 of the model group,nourishing-yin group,and huiyang group was significantly lower than that of the acute group(p<0.001),then there was no significant difference between the model group and nourishing-yin group(p>0.05),and the model group and nourishing-yin group were lower than that of the Huiyang group(p<0.001);on day 7,the expres sion of H19 in the model group and the nourishing-yin group was lower than that of the acute group(p<0.001),and there was no significant difference in the Huiyang group and the acute group(p>0.05).For the model group,the expression level of each group was higher than it(p<0.01),and the H19 expression level of the nourishing-yin group was lower than that of the Huiyang group(p<0.001);on the 10th day,the model The H19 expression in the yin-yang group,the yin-yang group,and the huiyang group was significantly lower than that of the acute group(p<0.001),the nourishing-yin group and the huiyang group were higher than the model group(p<0.05),and the expres sion of H19 in the nourishing-yin group was higher than that in the huiyang Group(p<0.001).Conclusion:1.Test Group wound healing is slower than the Control Group.2.External use of American Periplaneta extract and Huiyang Shengji Decoction can promote the healing of diabetic ulcer wounds,and the healing effect is mainly reflected after 5 days.3.The healing effect of Huiyang Shengji Decoction was more obvious than that of American Periplaneta extract in the middle stage,and there was no significant difference in the healing effect of the American Periplaneta extract and Huiyang Shengji Decoction in the later stage.4.The expres sion of lncRNA H19 will decrease in the case of hyperglycemia,but will increase after the application of Huiyang Shengji Decoction and American Periplaneta extract for external application,so Huiyang S hengji Decoction and American Periplaneta extract may play its role in promoting the healing of diabetic ulcer wounds through lncRNA H19.5.Although the expression of lncRNA H19 gradually decreased during the healing process of Huiyang Shengji Decoction,the expression level of the whole time period was significantly different from that of the model group.Therefore,Huiyang Shengji Decoction has anti-inflammatory properties and promotes cell proliferation and differentiation,Promotes the formation of collagen and granulation.6.The expression level of lnc RNA H19 increases continuously during the healing process of the American Periplaneta extract,which is significantly different from the model group in the middle and late stages.Therefore,the role of the American Periplaneta extract is mainly reflected in the promotion of collagen and granulation formation,and the reduction of scars. |
