Objective: To study the regulatory network between the specific expression of LncRNAs and mRNAs gene function of Huiyang Shengji Decoction on the wound surface after diabetic anal fistula operation,and explore the possible mechanism of Huiyang Shengji Decoction for wound healing after diabetic anal fistula operation.Methods: In the early stage,high-throughput transcriptome sequencing technology has been used to sequence all the collected diabetic anal fistula wound samples,and the difference between the two groups of lnc RNAs and mRNAs was screened within the range of|log2FC|>=1 and Pvalue<=0.05 differential expression.Expression profile,Venn diagram analysis and target gene prediction,and then select the lnc RNA related to this experiment from the target gene prediction for q-PCR verification,obtain meaningful positive indicators and correlate with differential mRNA to construct LncRNA-mRNA co-expression Based on the analysis of the LncRNA-mRNA co-expression network diagram,the function of the target LncRNA was explored from the genetic level to predict the possible mechanism of Huiyang Shengji Formula to promote wound healing after diabetic anal fistula.Results: Preliminary screening of LncRNAs and mRNAs with significant differential expression,Venn diagram analysis and target gene prediction,obtained 3 target genes,namely LncRNA(ENST00000450589,ENST00000596066 and ENST00000652702),and select the most functional LncRNA,namely ENST00000450589,for q-PCR Verification: After verification,the positive indicator ENST00000450589 is intersected with differential mRNA to obtain the LncRNA-mRNA co-expression network,and the analysis and prediction are based on the LncRNA-mRNA co-expression network.In the q-PCR verification,the expression level of ENST00000450589 in the experimental group was lower than that of the control group,which was consistent with the results of the high-throughput transcript data and played a down-regulation effect;in addition,ENST00000450589 cis-regulated target genes(KLHL20,SERPINC1,RC3H1,DARS2,ZBTB37 and CENPL).Among them,KLHL20,SERPINC1 and RC3H1 play a certain role in promoting angiogenesis,anti-inflammatory,immune regulation and other fields of healing,while the target genes DARS2,ZBTB37 and CENPL are rarely reported in inflammation and wound healing.Conclusion: Huiyang Shengji Recipe may promote the healing of diabetic anal fistula wounds by down-regulating the expression of ENST00000450589;it may also promote diabetes by cis-regulating target genes KLHL20,SERPINC1 and RC3H1 by ENST00000450589,affecting angiogenesis,anti-inflammatory response and regulating immune homeostasis Wound healing after anal fistula operation.
