Objective: In order to elucidate the mechanism of Taohong Siwu Decoction intervening the expression of Smad1 signal transduction protein to promote the repair of cartilage injury,the effects of Taohong Siwu Decoction on the expression of Smad1 signal transduction protein were observed by using modern molecular biological detection technology.Methods: All 40 healthy New Zealand white rabbits were randomly divided into four groups,namely Taohong Siwu Decoction group,glucosamine group,model group and normal control group,with 10 rabbits in each group.The models were made in Taohong Siwu Decoction group,glucosamine group and model group.The models of osteochondral injury,partial cartilage injury and full-thickness cartilage injury were made in the same rabbit’s left knee joint groove and femoral articular surface.On the fourth day after operation,the corresponding groups were given Taohong Siwu Decoction and glucosamine by gavage,the model group and the normal control group were given the same amount of normal saline by gavage,and 5 rabbits were randomly selected from each group at 4 weeks and 6 weeks after gavage After the incision,the injured cartilage was exposed for gross observation,and the articular surface including the lower end of femur was cut to detect the expression of Smad1 signal transduction protein for statistical analysis.Results:1.The average serum Smad1 concentration of Taohong Siwu Decoction group was significantly higher than that of glucosamine group and model group,but lower than that of control group(P < 0.05);2.The average gray value of WB detection in Taohong Siwu Decoction group was lower than that in the glucosamine group at the 4th week,but significantly higher than that in the model group,and higher than that in the glucosamine group at the 6th week.The control group was always higher than the other three groups(P < 0.05).3.There was no significant difference in the average serum Smad1 concentration between groups at the 4th and 6th week(P > 0.05)4.The average gray value of WB detection in the glucosamine group was significantly higher than that in the fourth week(P < 0.05),the difference was statistically significant,and the other three groups(P > 0.05),the difference was not statistically significantConclusion: Taohong Siwu Decoction can up regulate the expression of Smad1 signal transduction protein and promote the repair of cartilage injury,and the up regulation effect is better than that of glucosamine.
