Objective: To observe the effect of Chubi Jianxi Decoction on the expression of MMP-3,MMP-13 and Wnt/?-catenin signaling pathway protein in the serum of rats,and to explore the treatment of rat knee by Chubi Jianxi Decoction through Wnt/?-catenin signaling pathway The mechanism of action of joint osteoarthritis provides support and basis for the clinical treatment of KOA and the in-depth development and promotion of Chubi Jianxi Decoction.Methods: After feeding 70 male SPF SD rats adaptively for 1 week,50 rats were randomly selected for modeling,and 50 ul of sodium iodoacetate solution at a concentration of 40mg/ml was injected into the left knee joint of the rats.After modeling,they were randomly divided into model groups,Tenghuangjiangu tablets group,Chubijianxitang high,middle and low concentration group,10 rats in each group,and the remaining 20 rats were randomly divided into blank group and sham operation group with 10 rats in each group.To the left knee joint of rats in sham operation group 50 ul of normal saline was injected,and the blank group was not treated.After confirming that the model was successful,the samples were taken after2 weeks of intragastric intervention with different drugs.Observe the general conditions of the rat's diet,mental state,left knee joint swelling,joint flexion and extension,and activity;detect MMP-3 and MMP-13 in rat serum by double-antibody sandwich enzyme-linked immunosorbent assay(ELISA method)level changes;Western blot was used to detect the expression of Wnt-2 and ?-catenin protein.Results:1.The statistical analysis of the data of MMP-3 and MMP-13 in rat serum detected by ELISA shows that:comparing group F with group G,the level of MMP-3and MMP-13 in rat serum between group F and G can be obtained.No significant difference,p>0.05,no statistical significance;compared with the blank group,the levels of MMP-3 and MMP-13 in the model group,the experimental group with different concentrations of drugs,and the positive drug group were significantly increased,p<0.05,the difference It is statistically significant;compared with group E,the values of MMP-3 and MMP-13 in the serum of groups A,B,C,and D are all lower than group E,p<0.05,which is statistically significant;by comparing group A Compared with groups B,C,and D,it was found that the values of MMP-3 and MMP-13 in the serum of group A and group B were similar,p>0.05,no significant difference,no significant statistical significance;group A compared with group C,A The values of MMP-3 and MMP-13 in serum of group were lower than those of group C,p<0.05.Compared with group A,the values of MMP-3 and MMP-13 in serum of group D were significantly higher than those of group A,p<0.05,which is statistically significant;by comparing the levels of MMP-3 and MMP-13 in the serum of groups B,C,and D,it is found that the indicators of MMP-3 and MMP-13 in the serum have basically the same changing trend,that is,group B<C group<D group,group B compared with group C,p<0.05,group B compared with group D,p<0.05,group C compared with group D,p<0.05,the test analysis was statistically significant.2.Western blot was used to detect the expression of Wnt2 and ?-catenin protein in the cartilage of each group of rats.According to the statistical analysis of the data results,the statistical analysis showed that the sham operation group was compared with the blank group,p<0.05,and there was no significant change in the expression of Wnt2 and ?-catenin protein,No statistical significance;compared with the blank group,Wnt2,?-catenin protein expression increased in the model group,the experimental group with different concentrations of drugs,and the positive drug group,p<0.05,the difference was statistically significant;compared with the model group,The expression of Wnt2 and ?-catenin protein in groups A,B,C,and D decreased,p<0.05,which was statistically significant;by comparing group A with groups B,C,and D,group A and group B Wnt2,?-catenin The protein expression is similar,p>0.05,which is not statistically significant;by comparing the Wnt2 and?-catenin protein expression in the groups B,C,and D,it is found that the protein expression indicators have basically the same changing trend,that is,group B<group C<group D,Group B compared with group C,p<0.05,group B compared with group D,p<0.05,group C compared with group D,p<0.05,the test analysis was statistically significant.Conclusion: Chubi Jianxi Decoction can inhibit the expression of Wnt2 and ?-catenin protein in chondrocytes in the KOA rat model,thereby inhibiting the Wnt/?-catenin signaling pathway and down-regulating the expression of the upstream and downstream target genes MMP-3 and MMP-13 in the signaling pathway,Reduce its catabolism to articular cartilage,and achieve the purpose of treating knee osteoarthritis.
