Objective: The aim of this study was to clarify the expression of NLRP3 inflammasome and its related pathways in the model of acne inflammation,to clarify the mechanism of pomegranate peel polyphenol cream i nhibiting NLRP3 inflammasome anti-inflammatory and anti-acne,and to determine the optimal concentration of NLRP3 inflammasome for the treatment of acne.Methods: Forty-two male and female SPF SD rats were randomly divided into blank group,model group,ma trix group,pomegranate peel polyphenols low,medium and high concentration group,and clindamycin group,with 6rats in each group.Except for the blank group,the other groups were given the auricle acne model of rats.The visual observation and patholog ical results indicated that after the model was successfully established,the auricle of rats in each group was given topical drugs according to the test factors.The matrix group was given 0.5g of matrix cream,the pomegranate peel polyphenol group was gi ven 0.5g of pomegranate peel polyphenol cream of different concentrations,and the clindamycin group was given0.5g of clindamycin hydrochloride cream.The blank group and model group were treated with the same amount of distilled water twice a day for 2weeks,and samples were collected 24 hours after the last treatment.The changes of auricle skin lesions were observed by naked eyes.Histopathological changes were observed by HE staining.The expressions of NLRP3,Caspase-1,ASC and MMP-2 in the skin lesions were detected by immunohistochemistry.Results:1.Macroscopic observation: Compared with the blank group,the auricle skin lesions in the model group and the matrix group showed severe inflammation;Compared with the model group,the auricle skin lesi ons of rats in pomegranate peel polyphenol groups and clindamycin groups were improved to varying degrees,and the medium concentration group was the most significant.2.Histopathological observation: Compared with the blank group,the number of sebaceous glands in the auricular skin lesions in the model group and the matrix group was increased,the infiltration of inflammatory cells was obvious,and local bleeding was visible;Compared with model group,inflammatory cell infiltration and sebaceous gland h yperplasia were reduced in pomegranate peel polyphenol group and clindamycin group.3.Immunohistochemical analysis showed that the expressions of NLRP3,ASC,Caspase-1 and MMP-2 in auricle skin lesions of model group were significantly increased compared with blank group(P < 0.05);Compared with model group,NLRP3 expression was significantly decreased in pomegranate peel polyphenol groups and clindamycin groups(P < 0.05),and the decrease was most significant in medium concentration group(P <0.05).ASC expression in pomegranate skin polyphenols medium and high concentration groups decreased(P < 0.05),but there was no significant difference between low concentration group and clindamycin group(P >0.05).Caspase-1 expression was decreased in pomegran ate peel polyphenol groups and clindamycin groups(P < 0.05),and the decrease was the most significant in pomegranate peel polyphenol group with medium concentration.The expression of MMP-2 in pomegranate skin polyphenol low and high concentration groups was decreased(P < 0.05),but there was no significant difference in the expression of MMP-2 in medium concentration group(P > 0.05),and the expression of MMP-2 in clindamycin group was increased(P < 0.05).4.ELISA test showed that compared with blank group,the levels of inflammatory cytokines IL-1β and IL-18 in serum samples of model group were significantly increased(P < 0.05);Compared with model group,pomegranate peel polyphenol groups and clindamycin groups were significantly decreased(P < 0.05);Compared with different concentration of pomegranate peel polyphenol groups,the decrease of high concentration group was the most significant,followed by medium concentration group.Conclusion:1.Topical application of pomegranate peel polyphenol cr eam can significantly improve the inflammatory response of the rat acne model.2.Pomegranate peel polyphenol cream can achieve anti-inflammatory and anti-acne effects by inhibiting NLRP3 inflammatory corpuses.3.Pomegranate peel polyphenols in medium and high concentration groups can significantly inhibit the immune response of acne inflammation.
