Objective: Alzheimer's disease,as an aging disease,is harm to the public hygiene.Aerobic exercise has a good effect as a means to delay and improve AD.In this study,APP/PS1 double transgenic mice were used to intervene with appropriate aerobic exercise for a long time.From the perspective of mitochondrial autophagy,the theory of preventing AD was discussed.Methods:(1)20 3-month C57/B6 L wild mice were divided into two groups:non-exercise group(WR)and exercise group(WE).20 3-month APP/PS1 transgenic mice were divided into two groups: non-exercise group(TR)and exercise group(TE).(2)TE group and WE group for 20 weeks of treadmill training,WR group and The TR group was fed naturally for 20 weeks.After 20 weeks of aerobic exercise,all four groups of mice underwent behavioral tests simultaneously.(3)break out the frontal lobe,using Western blot to determine A?,LC3-?/ LC3-?,DRP1,Mfn2 and PINK1,Parkin protein content,with immunofluorescence experiments in mice in the frontal lobe Parkin and PINK1 positioning for fluorescence.Results:(1)In the Morris water maze,the incubation time and total swimming distance of TR group were greater than those of WR group,while TE group were smaller than those of TR group.The platform time and distance percentage in TR group were lower than those in WR group.the TE group were all higher than those in the TR group.Space exploration experiment: the number of platform crossing,platform quadrant time and distance percentage in TR group were less than those in WR group;The TE group were higher than those in TR group(P < 0.05).(2)In the eight-arm maze experiment,the working memory error frequency,reference memory error frequency(P < 0.05)and work completion time in TR group were all higher than those in WR group,while in TE group were all lower than those in TR group(P< 0.05).(3)TR group A?,DRP1,PINK1,Parkin protein content than the WR group conspicuously raise(P < 0.05),while those in TE group were decreased compared with TR group,with significant differences(P < 0.05).TR group compared with WR,LC3-/ LC3 ?-(? P < 0.05),Mfn2 protein are falling;The protein contents of TE group were increased compared with TR group,with significant differences(P < 0.05,P < 0.01).(4)Immunofluorescence showed that the expression of PINK1 and Parkin increased in TR group compared with WR group.Compared with TR group,TE group showed reduced expression.Conclusion: 20 weeks of treadmill aerobic training can reduce the division of frontal lobe mitochondria of AD mice,promote fusion,regulate autophagy,improve the pathological and behavioral characteristics,improve its learning ability and memory performance,and prevent the occurrence of AD.
