| Objective: To use Huanglian Wendan Decoction to intervene with traditional Chinese medicine in type 2 diabetes(T2DM)male mice,and to preliminarily explore the effect of Huanglian Wendan Decoction on liver glucose and lipid metabolism in T2 DM mice based on PI3K/Akt/Fox O1 signaling pathway;to explore the prevention and treatment effects of Huanglian Wendan Decoction on T2 DM and related mechanisms;Provide theoretical and experimental data support for the promotion of Huanglian Wendan Decoction in the treatment of diabetes and its complications in the future.Methods: Feeding with high-fat and high-sugar feed combined with intraperitoneal injection of small dose of streptozotocin(STZ)to induce the establishment of a mouse T2 DM model.The experiment set up a blank group,a control group and a traditional Chinese medicine group to investigate the effects of Huanglian Wendan Decoction on fasting blood glucose in mice(FBG),glucose tolerance(IPGTT),C peptide,body mass(W),liver index,liver tissue steatosis,and the influence of PI3 K,AKT,Fox O1 protein expression.result:1.FBG: In the 13 th week,compared with the blank group,the FBG of the mice in the control group was extremely significantly increased(P<0.001);compared with the control group,the FBG of the mice in the traditional Chinese medicine group was extremely significantly lower(P<0.001).2.IPGTT: Compared with the blank group,the area under the blood glucose-time curve(AUC)of the pre-administration group and the post-administration control group after model building was extremely significant(P<0.001);After the treatment,the AUC of the Chinese medicine group was extremely significantly(P<0.001)reduced.3.W: In the 9th week(before administration)and 13 th week,compared with the blank group,the W of the mice in the control group increased significantly(P<0.001);at the 9th week(before administration),Compared with the blank group,the W of the mice in the Chinese medicine group was extremely significantly increased(P<0.001);at the 13 th week,compared with the control group,the W of the Chinese medicine group was extremely significantly increased(P<0.001);W changed,compared with the control group Compared with the traditional Chinese medicine group,it was significantly higher(P<0.01).4.Liver index: Compared with the blank group,the liver index of the mice in the control group increased significantly;compared with the control group,the liver index of the mice in the traditional Chinese medicine group decreased significantly.5.HE staining of liver tissue: the control group showed moderate-severe hepatocyte steatosis,and the traditional Chinese medicine group showed mild-moderate steatosis.Compared with the control group,the liver cells of the traditional Chinese medicine group were arranged more neatly,the number of fatty liver cells decreased,and the cytoplasm Lipid droplets are significantly reduced.6.Target protein expression: Compared with the blank group,PI3 k and Akt in the control group were significantly reduced(p<0.01),and Fox O1 was significantly increased(p<0.05);compared with the control group,PI3 k and Akt in the traditional Chinese medicine group were significantly increased(p<0.01),Fox O1 was significantly reduced(p<0.05).Conclusion: Huanglian Wendan Decoction has the effect of lowering blood sugar and can significantly reduce the fasting blood glucose of T2 DM mice;significantly improve the glucose tolerance and IR of T2 DM mice;significantly improve the "weight loss" of T2 DM clinical manifestations;improve the liver of mice Fatty changes in tissues of liver cells can reduce liver coefficient;it can activate the liver PI3K/AKT signaling pathway,which may be achieved by regulating the liver’s PI3K/AKT/Fox O1 pathway to reduce blood sugar,improve IR,and regulate lipid metabolism. |
