Objective:To observe the effects of Huanglian Wendan Decoction on diet-induced insulin resistance in rats with metabolic syndrome and aortic H dyeing,serum vWF,parallel to explore its mechanism.Method:Sixty 7-week-old healthy male SD rats were randomly divi -ded into 2 groups which were vacuity group(n=10)and experimental gro -up(n=50).Experimental group was fed with high Iipid,high glucose and high salt diet for 20 weeks to induce experimental metabolic syndrome. 37 SD rats in experimental group which were modeled successfully were randomly divided into 4 groups:model group,huanglian wendan decoction high-dose group,low-dose group and Metformin Hydrochioride control group,which were lavaged with the appropriate drug for 4 weeks.To observe the general state of rats,dynamic state measure weight,blood pressu -re,lipids,fasting blood glucose,serum insulin,and other indicators,calcul -ate Lee's index and HOMA-IR,to evaluate model and drug effect.Before and after drug therapy,we observed the pathological change of aorta by H staining method,and tested the vWF levels of serum by ELISA metho -d.Result:1.After fed for 20 weeks,the weight,abdominal circumference,blood pressure,lipids TG,Tcand,LDL,fasting glucose,fasting insulin,HOMA-IR of rats model with MS were significantly higher,HDLC level of reducti-on(P<0.01),compared with the vacuity group.2.FBG,FINS,HOMA-IR of Model group were significantly higher tha -n the vacuity group(P<0.01).The weight,Lee's index,visceral fat weight, visceral fat and body weight ratio,FBG,FINS,HOMA-IR in metformin gro -up and huanglian high-group were decreased(P<0.01orP<0.01),compared with model group,treatment effects are similar to the two groups,the diff -erence was not significant(P> 0.05).The blood pressure,lipids TG,TC, LDLC were significantly decreased(P<0.01)in huanglian high-group.The lipids TG,TC,LDLC and HDLC were not significantly changed in metfor-min group(P> 0.05).The weight and lipids TC were significantly lower in huanglian low-group(P> 0.05).There were all compared with the mode -1 group.3.Aortic H staining showed:Model group had slightly thickened int -ima and moderate hyperplasia of endothelial cells compared with the vacuity group.Smooth Endometrial and significantly inhibited proliferatio -n are in huanglian high-group.Huanglian wendan low-dose group had varying degrees of intimal thickening,Metformin group was of no mild intimal thickening.4.Compared with the vacuity group,the serum vWF levels of MS model group were significantly higher(P<0.01).Compared with the MS model,the serum vWF in all treatment groups were significantly lower (P<0.01).Conclusion:1.The weight,blood pressure,abdominal obesity,insulin resistance and disordered lipid metabolism of MS rats can be improved by Huanglian Wendan Decoction.2.A wide range of the IR and vascular endothelial dysfunction wer-e existed in rats with metabolic syndrome.3.Huanglian Wendan Decoction can improve the MS rats aortic pathology structural changes of H staining,lower serum vWF,with the pro -tection of vascular endothelial cells.
