Objective:Hymenocallis littoralis is a perennial herb of Amarylliaceae.It has been used for the treatment of relaxing tendons,promoting blood circulation,reducing swelling and relieving pain.Alkaloids are the main chemical constituents in H.littoralis.Our LC-MS analysis suggested that,in addition to the alkaloid components,there are more other types of components in H.littoralis,indicating that the chemical composition of the non-alkaloid components H.littoralis is worth exploring.In view of this,this subject will conduct a systematic study on the chemical constituents of H.littoralis,and it is hoped that potential active compounds will be discovered through activity screening.Methods:The whole plant is extracted with 95%ethanol,and the solvent extraction is used for crude separation,and then a comprehensive use of chromatographic separation methods,such as silica gel column chromatography,gel column chromatography,MCI column chromatography,and preparative HPLC,etc.The chemical components of H.littoralis are systematically stuied.The structures of the isolated compounds were identified by methods such as physical and chemical properties determination,HR-ESI-MS,1H-NMR,13C-NMR,1H-1HCOSY,HSQC,HMBC,and CD.The isolated compounds are screened for promoting wound healing activity by promoting mouse fibroblast proliferation experiments,Meanwhile,the isolated compounds are screened for cytotoxic activity through lung cancer cell proliferation inhibition experiments.Results:Twenty seven compounds were isolated and identified as(2S,5"R)-7,4’-dihydroxy-8-methyl-6-(2"-oxopyrrolidin-5"-yl)-flavanone(1),(2R,"S)-7,4’-dihydroxy-8-methyl-6-(2"-oxopyrrolidin-5"-yl)-flavanone(2),5,7-dihydroxy-6,8-dimethoxy-2-(hydroxymethyl)-4Hchromen-4-one(3),myrsinionoside A(4),undulatoside A(5),(2S)-4,7-dihydroxyflavane(6),naringenin(7),4’,7-hydroxy-8-met hylflavanone(8),8-methylnaringenin(9),8-deMethylrhododendron(10),6-methyl-aromadendrin(11),5,7,4’-trihydroxy-8-methylflavone(12),syzalterin(13),5,7,4-trihydroxy-6-methylflavone(14),isoglycyrrhizi cacid(15),2’,4,4’-trihydroxy-3’-methylchalcone(16),undatusidesC(17),Phenyl ethyl-O-[β-D-xylopyranosyl-(1→2)-β-D-glucopyranoside](18),4-methyl eucomate(19),4-methyl eucomate(20),hydroxyeucomic acid(21),pancratistein(22),narciclasine-4-O-β-D-Glucopyranoside(23),lycorine(24),11-Hydroxyvittatine(25),Tazettine(26),β-adenosine(27).Among them,compounds 1,2,3 are new compounds,and 19 compounds 4-21,23 and 27 are isolated from H.Littoralis for the first time.At the same time,a series of compounds isolated from H.Littoralis were tested wound healing activities through L929 cells.Compared with the blank group,the administration group had no effect on the proliferation of L929 cells.However,compounds 1,9,12 and 15 showed obvious cytotoxicity.What’s more,those compounds were tested cytotoxicity activities against A549 cells.Compared with the blank group,compounds 5,6,8,10,12 and 14 showed obvious cytotoxicity.Conclusion:In this paper,27 compounds were isolated,and flavonoids,alkaloids and phenylpropanoids are the main chemical constituents.Among them,compounds 1,2 and 3 are three new compounds,and other 19 compounds are isolated from H.Littoralis for the first time.Specific categories include flavonoid alkaloids(1,2),narciclasine type alkaloids(22,23),lycorine type alkaloid(24),crinine type alkaloid(25),tazettine type alkaloid(26),and other types of alkaloids(27),chromoproketones(5,7),flavane(6),dihydroflavones(7,8,9,10),dihydroflavonol(11),flavonoids(12,13,14),chalcones(15,16),phenylpropanoids(19,20,21)and other compounds(4,17,18).Some isolated compounds showed significant cytotoxic activity of lung cancer cells.The results obtained in this study are helpful for a more comprehensive understanding of the composition of the chemical components of H.littoralis,enrich the research content of the chemical composition of H.littoralis and their biological activity,and provide a comparison for the in-depth study of the active chemical components in the H.littoralis.
