| Objective:The mechanism of baicalein action on triple negative breast cancer was analyzed by bioinformatics combined with network pharmacology.The target of baicalein action on the core gene of triple negative breast cancer was simulated by molecular docking method,and the analysis results were verified by in vitro cell experiments.To prove the theoretical basis of the application of natural small molecule baicalin in triple negative breast cancer.Methods: Human triple negative breast cancer gene chip was searched by GEO database.Using the GEO2 R online processing tool,the chips were divided into normal group and tumor group for online analysis.According to adj.P.V al and | log FC | screening,the result of the processing map Wayne,through online tools of differentially expressed genes,and get a different chip through a STRING of online data analysis after an analysis of the topology of differentially expressed genes,import Cytoscape core gene screening according to the Degree value,GO and KEGG enrichment analysis of core genes was performed by DAVID database.The effective components of Scutellaria baicalensis were searched by TCMSP,and the relevant effective components of Scutellaria baicalensis were selected as the research target.The minimum free energy was calculated by molecular docking to simulate the binding site of baicalin to core genes.The effect of baicalein on the biological activity of breast cancer cells was tested in vitro.The effects of different concentrations of baicalein on the activity of breast cancer cells were tested by cell activity assay to screen out the optimal dose.The inhibitory effect of baicalin on the migration of breast cancer cells was tested by scratch test.The inhibitory effect of baicalein on breast cancer invasion was verified by Transwell invasion assay.Apoptosis assay was used to verify whether baicalein can promote the apoptosis of breast cancer cells.Cell cycle assay was performed to determine the effect of baicalein on cell cycle progression of breast cancer.PCR and Western Blot were used to test whether baicalein inhibited the growth of breast cancer cells by inhibiting the expression of core genes.Results: There were 613 common differentially expressed genes of GSE45827,GSE61724 and GSE38959,among which 10 core genes were obtained through topological analysis.CDK1,PLK1,CCNA2,BUB1,CCNB1,KIF11,AURKA,TOP2 A,BUB1B,MAD2L1.According to the results of enrichment analysis,it was found that the core genes were mainly enriched in the cell cycle pathway,in which CDK1 and CCNB1 jointly affect PLK1.Baicalein was screened by TCMSP and ADME as the research component of Scutellaria baicalein.By online Wayne tool,it was found that the common gene of baicalein corresponding target and core differentially expressed genes in breast cancer was PLK1,and CCNB1 and CDK1 combined in the KEGG pathway jointly affected PLK1.Therefore,PLK1,CCNB1 and CDK1 are the core genes.Through activity test,scratch test,migration test,apoptosis test and cell cycle test,it was found that baicalin could inhibit the proliferation and migration ability of breast cancer cells,promote the apoptosis of breast cancer cells,and cause the arrest of breast cancer cell cycle.PCR and Western Blot experiments showed that baicalein inhibited the expression of PLK1,CCNB1 and CDK1 in breast cancer by dose compared with the blank control group.Conclusion: Triple negative breast cancer is a serious threat to women’s life and health because of the characteristics of single treatment and early onset age.Baicalein,as a small molecule active component of Scutellaria baicalensis,can inhibit the proliferation and migration of breast cancer cells.Promote apoptosis and other ways to inhibit tumor growth.By inhibiting the expression of PLK1,CDK1 and CCNB1,breast cancer cell cycle is affected and tumor cell growth is eventually inhibited.It explained that baicalein inhibits the growth of tumor cells through multiple targets,and provided the theory for clinical application of baicalein as a complementary therapy for triple negative breast cancer. |
