| Snow chrysanthemum(Coreopsis tinctoria Nutt.),black-leaved apricot(Armeniaca vulgaris Lam.),and Ruoqiang jujube(Elaeagnus crispa Thunb.)are the three endemic species in Xinjiang,which are rich in flavonoids,polyphenols and polysaccharides.However,the current food processing field has not fully exploited the resources of the above three specialties,which restricted the further upgrading and development of the local industry.Therefore,this topic used the above three as raw materials to develop compound drinks with anti-oxidation and anti-fatigue related health care functions.Response surface methodology was used to optimize the optimal extraction process of total flavonoids from snow chrysanthemum,total polyphenols from dried apricot and total polysaccharide from jujube.And then the formula of compound drink was obtained by orthogonal test with sensory score as index.Finally,in vitro anti-oxidation and anti-fatigue experiments were carried out to prepare a compound drink with good taste and health function.The main research contents of this paper were as follows:(1)Using flavonoid,polyphenol and polysaccharide as index respectively,single factor and Box Behnken response surface were used sequentially to study the extraction technology of snow chrysanthemum,dried apricot and jujube respectively.The optimal extraction process of snow chrysanthemum was as follows:solid-liquid ratio 1:90,extraction time 23 min,temperature 95℃,extraction twice,the total extraction amount of flavonoids was 316.25 mg/g under this condition.The optimal extraction process of dried apricot was as follows:solid-liquid ratio 1:7,time 2 h,temperature 90℃,extraction twice,the total polyphenol extraction amount was 3.05 mg/g under this condition.The optimal extraction process of Ruoqiang jujube was as follows:solid-liquid ratio 1:18,time 88 min,temperature 73℃,extraction twice,the total polysaccharide extraction yield was 667.76 mg/g under this condition.(2)The compound beverage was prepared with the extracts of snow chrysanthemum,dried apricot and jujube as the main additives,and also supplemented with proper amount of sucrose and citric acid.On the basis of single factor experiment and then L9(34)three-factor and three-level orthogonal experiment,the optimal technology of compound beverage was determined as follows:snow chrysanthemum-dried apricot juice:jujube juice(V/V)1:1,flavoring agent:sucrose 5%,with a citric acid of 0.1%.The physicochemical indexes and microbial content of the beverage were determined.The total acid of the composite drink was 1.45 g/L,the soluble solid was 8.2%,the total number of colonies,coli group and mold and yeast were not detected.Antioxidant test showed that DPPH scavenging activity of the beverage was 825.24μg Trolox/m L.The total antioxidant capacity was 90.32 U/m L.Acute toxicity test was performed on a compound beverage of Coreopsis tinctoria to evaluate the safety of it,which dose of mice by gavage was 40m L/kg,the same amount of distilled water in control group was gavaged.No obvious symptoms of poisoning were observed and no animal died during the observation period two weeks after administration.Therefore,the maximum dose of drug was 30 g/kg.d(42 g crude drug/kg.d),which was equivalent to 48 times of 60 kg per day for adults.According to the acute toxicity grading standard,the sample was preliminarily judged to be nontoxic.(3)Through network pharmacology,the mechanism of action of a compound beverage of Coreopsis tinctoria to relieve exercise fatigue was studied,and the results showed that quercetin,Butein,Okanin mainly acted on targets such as EGFR、MAPK3、MAPK1和ESR1,and were regulated by PI3K-AKT、HIF-1 and MAPK signaling pathways to relieve exercise fatigue.The weight-bearing swimming test in mice further revealed the antifatigue effect of the compound beverage. |
