Abstract/Summary:
Euphorbia, covering about 2,000 species,is the largest genus in angiosperms.According to literatures,Euphorbia plants are rich in natural active ingredients and possess the diverse skeletons and bioactivity, which making this genus as an important resource for the discovery of drug lead compounds.Euphorbia fischeriana Steud,is a kind of perennial herbaceous plant belonging to Euphorbia of family Euphorbiaceae.E.fischeriana Steud was first reported in the "Shennong Materia Medica" to have bitter taste, pungence, high toxicity, with the effect of eliminating phlegm, removing blisters and killing insecticidal. In recent years, researchs on E.fischeriana Steud have also focused on chemical constituents with anti-tumor and anti-tuberculosis and a series of pharmacologically active substances have been isolated, which has attracted widespread attention. So far,the research on the chemical composition of E.fischeriana collected from Shangri-La in Yunnan Province has little been reported.E.hypericifolia,an annual herbaceous plant in the Euphorbia of Euphorbiaceae family, has the effect of clearing heat and moisture, astringent, and antipruritic, and can be used to treat diarrhea and itchy skin diseases. So far, only 3 papers about the chemical component of this species have been reported. Therefore, the above two plants were selected for chemical composition research to provide a material basis for subsequent biological activity screening. This thesis consists of three chapters:Chapter 1 summarized the atisane-type diterpenoids isolated from Euphorbia plants since 1990 and their biological activity. Also the details of structure, name, plant origin of the them were discussed.Chapter 2 dealed with the chemical compositions of ethyl acetate part of 70%acetone aqueous extract from E.fischeriana steud. In this paper,a variety of modern separation methods including normal and reverse silica gel column chromatography,MCI,dextran gel column chromatography and semi-preparative high performance liquid chromatography were used to isolate 18 compounds. The structure types of these compounds covers diterpenes, triterpenes, flavonoids, and coumarins,in which compounds 1、9 and 10 were new ones, and compounds 1 ~ 18 were isolated from this plant for the first time.Chapter 3 discusses the chemical composition of petroleum ether and ethyl acetate parts of 70% acetone aqueous extract of E.hypericifolia.In this thesis, various separation technologies were used and a total of 82 compounds were obtained.Their structural types involves diterpenes, triterpenes, steroids, flavonoids, emodin, coumarin, etc.,among which compounds 1 and 29 were new compounds, and compounds 1 ~ 69、71 ~ 79、81 ~ 82 were firstly isolated from this plant.
