| Tea fungus is a functional beverage which is mainly fermented by various microorganisms such as acetic acid bacteria,yeasts and lactic acid bacteria with tea sugar as the fermentation substrate,tea fungus includes biofilm,fungus broth and other substances..Studies have shown that tea fungus broth has a variety of probiotic effects,such as anti-oxidation,anti-aging,anti-inflammatory,and antibacterial.In this study,Drosophila melanogaster was used as an animal model to explore the effects of unfermented black tea and different density of tea fungus broth on the lifespan,reproduction and antioxidant capacity of D.melanogaster.In addition,this study also evaluated the effects of unfermented black tea and tea fungus broth on the intestinal flora of D.melanogaster.The main findings are as follows:(1)Adding tea fungus broth(KT1,10 m L/L;KT2,20 m L/L;KT3,30 m L/L)significantly prolonged the lifespan of male D.melanogaster and the KT3 group significantly prolonged the lifespan of female D.melanogaster.Significantly improve the ingestion capacity of D.melanogaster so as to increase the average body weight and body length of D.melanogaster,and shorten the developmental period and pupal stage of D.melanogaster.In the meanwhile,the crawling ability of D.melanogaster and the ability to resist hunger,cold and heat stress were also enhanced.The best improvement of each index was shown in group KT3,and such improvement increased as the feeding time expands.(2)As was shown in the research about the effect of black tea and tea fungus broth on D.melanogaster reproduction,the mating rate of D.melanogaster was improved in group BT and group KT.The mating latency of D.melanogaster was significantly shortened in group KT3,and the mating duration and fecundity of D.melanogaster were promoted dramatically as well.BT and KT3 group demonstrated the promotion of the ovarian development of females D.melanogaster.(3)Black tea and tea fungus broth could increase the activities of CAT,T-SOD and T-AOC in parental and third-generation D.melanogaster,and such increase would be promoted over time.The KT3 group could significant reduction of the MDA content in parental and third-generation D.melanogaster and the decline continued to increase over time.(4)After the D.melanogaster ingested the black tea(G.BT),the diversity of intestinal microbes increased significantly.Compared with the control group(G.CK),the relative abundance of Proteobacteria decreased and the relative abundance of Firmicutes increased.The relative abundance of Acetobacter decreased and the relative abundance of Lactobacillus increased.After ingesting the tea fungus broth(G.KT3),the abundance of intestinal microorganisms increased,and the change in diversity was not remarkable.The relative abundances of Firmicutes,Acetobacteriaceae,Rhizobiaceae and Acetobacter were increased.The metabolic genes of cell growth and death in group G.BT were prominently increased,and the relative abundance of metabolic genes in Amino acid metabolism,Viral protein family and Environmental adaptation in group G.KT3 were markedly enhanced.This study shows that tea fungus broth can promote the lifespan,growth and development,reproduction,exercise ability and antioxidant capacity of D.melanogaster,but the effect of tea fungus broth on the intestinal flora of D.melanogaste is not significant.Our research would provide theoretical basis for the utilization of tea fungus broth and the development of new health care products made from tea fungus broth. |
