In this study,Dendrobium officinale was used as experimental material,and the physicochemical properties of the shell material were preliminarily studied.The harmless fermentation scheme of the shellfish substrate was discussed in depth.Finally,the growth characteristics and nutrient contents of Dendrobium officinale were studied systematically.The main findings are as follows:1.Dendrobium officinale is suitable for growth in a weakly acidic environment,and the base of the shellfish is highly alkaline and cannot be directly used as a cultivation substrate for Dendrobium officinale.Therefore,by adding sawdust and sheep manure fermentation,the alkalinity of the shellfish matrix is reduced.Harmful treatment.2.The survival rate of Dendrobium officinale in each cultivated substrate was calculated after 240 days of transplanting.It was found that the survival rate of Dendrobium officinalewas higher than that of common cultivated pine bark and coconut shell,indicating that the shellfish matrix is very ideal.Dendrobium officinale cultivation substrate.The morphological indexes of Dendrobium officinale were determined by measuring the different ratios of the crustaceous substrate and the common substrate pine bark and coconut shell.The results showed that the mixed matrix of the 4th shell:sawdust:sheep manure=6:3:1 and 5 mixed matrix Shell:Sawdust:Sheep manure=5:4:1 is the best ratio of the shell material.3.The phenol-sulfuric acid method was used to detect the polysaccharide content of Dendrobium officinale.The regression equation of glucose standard solution was:Y=0.0658X+0.0423,R2=0.999.The linear relationship is good between 2.570 μg/mL and 12.860 μg/mL.The method validation results show that the detection method is reliable.The polysaccharide content of each sample ranged from 14.93%to 21.95%,and the order of polysaccharide content was:matrix 4>matrix 5>matrix 7>matrix 3>matrix 2>matrix 1>matrix 8.4.HPLC method was established to detect the peak area ratio of mannose and glucose in Dendrobium officinale.The optimized conditions were as follows:column:mobile phase:acetonitrile-0.02 mol/L ammonium acetate(18:82);Flow rate:1 ml/min;column temperature:30℃;detection wavelength:250 nm;injection amount:10 μL,methodological verification results show that the detection method is reliable.The peak area ratio of mannose to glucose in each sample ranged from 2.15 to 2.68,and the peak areas of mannose and glucose were ranked as follows:matrix 4>matrix 5>matrix 7>matrix 3>matrix 2>matrix l>matrix 8.5.The quality indexes of the polysaccharide content of Dendrobium officinale and the peak area ratio of mannose and glucose were compared.The results showed that the mixed matrix of No.4 was:Sawdust:Sheep manure=6:3:1 and No.5 mixed matrix Pu shell:sawdust:sheep manure=5:4:1 is a high quality cultivation substrate of Dendrobium officinale.
