| The Wheat albinism line is an albino mutant of wild-type Wheat Aibian 1.There is no obvious phenotypic difference between these two wheat lines at the seedling stage,but when the temperature continues to decrease,the stage albino begins from the young leaves.As the temperature gradually increases,the albino part can be green again.Previous studies have shown that the albino property is regulated by both nuclear and cytoplasm factors,low temperature is a necessary condition for albino,but the mechanism of albino has not been fully elucidated.In this thesis,the transcriptome and proteome expression profiles of two materials at room temperature and low temperature were established using high-throughput experimental techniques.Genes and proteins that are induced by low temperature and related with albino were further mined.Moreover,the potential molecular mechanism of the albino process of albinism line was explored.The analysis results are as follows:1.Low temperature stress induces a significant difference in the expression of 32,801 genes between the Wheat albinism line and Wheat Aibian 1.These differentially expressed genes are involved in protein phosphorylation,biological synthesis of cell walls,maintenance of plant photosystem II stability,synthesis of polysaccharides,and transport of divalent metal ions.2.At the protein level,there were 431 differentially expressed proteins between the Wheat albinism line and Wheat Aibian 1 at room temperature.The genes corresponding to these proteins are involved in the biological processes associated with photosynthesis.3.There are 96 genes between the Wheat albinism line and Wheat Aibian 1,which are significantly different in transcriptional level and protein level,involving chlorophyll synthesis and low temperature stress response.4.According to the results of transcriptome,proteome,and PCR validation,three genes with significant differences in expression between the Wheat albinism line and Wheat Aibian1 were obtained.Of these,WCOR15-2D and Dna J11-7B were significantly down-regulated in the albinism line,SGR-2D was significantly up-regulated in the albinism line.The correlation between these three genes and the albino phenomenon deserves further study. |
