Soybean[Glycine max(L.)Merr.],is one of the most important crops in the country and in our world.Water-soluble protein is one of the most abundant soybean proteins,accounting for about 70%of the protein content of soybean,which is an important source of good protein intake.The water-soluble protein is a typical quantitative character,which is influenced by environment and heredity.For soybean grain of different varieties,different growth environment,the differences in soybean seed water-soluble protein is endless also and same,and the ratio of water soluble protein content and protein content can reflect the storage life of soybean grain,thus expanded soybean grain related properties such as water soluble protein content of QTL mapping research,on the agricultural production,high protein soybean varieties of breeding with excellent quality and improve the practical value and storage life of soybean has a positive and important meaning.This research use high water-soluble protein material Guizao1 and low water soluble protein content material BX13,to construct recombinant inbred lines,as test materials,in three environments(different years and different regions)planting respectively,planting specifications according to the national regional test standard planting,spaced 50cm,planting distance 10cm,three repeat,mixed harvest,uniform sampling,the agricultural industry standard of the People’s Republic of China 2006 kjeldahl nitrogen determination method of water soluble protein content of phenotype data are collected,analysis of phenotype data at the same time.The high density genetic linkage map and complex interval mapping were used to map the water-soluble protein content of soybean seeds,which laid a certain foundation for the study of water-soluble protein content of soybean.The following are the main research conclusions:The correlation analysis of water-soluble protein content and protein content in three different environment showed that the water-soluble protein content and protein content were positively correlated,with a correlation coefficient r=0.202(p<0.001),from the low correlation coefficient,we can know that the varieties with high water-soluble protein content may not have high protein content.The ratio of water-soluble protein content and water-soluble protein content to protein content was significantly positive,with the correlation coefficient r=0.9952(p<0.001),which could indirectly reflect the storage life of soybean seeds according to the content of water-soluble protein.There was no correlation between the content of water-soluble protein and the content of oil.The resequencing genotypes of Guizao1×BX13(GB13)population were combined with Win QTLChart2.5 to accurately locate the phenotype data of water-soluble protein content by composite interval mapping(CIM).A total of 11 QTLS were detected,with LOD values ranging from 2.71 to 10.14,and phenotypic variance interpretation rate(R~2)ranging from 4.26%to 17.28%.There were 1 QTL were found on chr4,2 QTLs were found on chr8,4 QTLs were found on chr13 and 4 QTLs were found on chr20.Among them,2 principal effect QTLs were consistent with those determined by predecessors,and 3 principal effect QTLs were determined to 6 new principal effect QTLs near the results of previous localization.By searching the genes in the main QTLs interval in the database,118 genes were found on chr4,180 genes were found on chr8,396 genes were found on chr13 and 954 genes were found on chr20.Through Gene Ontology(GO)enrichment classification of these genes and annotation analysis of Phytozome database genes,a total of 15 candidate genes related to water-soluble protein content,including Glyma.04G044700,Glyma.04G045700,Glyma.08G199200,Glyma.08G199300,Glyma.08G201000,Glyma.13G097400,Glyma.13G106300,Glyma.13G107300,Glyma.13G109200,Glyma.20G197500,Glyma.20G221400,Glyma.20G100500,Glyma.20G102500,Glyma.20G069300,Glyma.20G080900,the functional verification of 15 candidate genes remains to be further identified.To sum up,this study provides a reference for obtaining varieties with high water-soluble protein content,and also provides a certain reference for studying QTLs localization of water-soluble protein content in grains.
