"Zijuan"(Camellia sinensis cv.Zijuan)is a special tea tree germplasm in Yunnan Province,with the characteristics of purple bud,purple leaf and purple stem.Anthocyanins are the main characteristic components of purple leaves in Zijuan tea.Anthocyanins are more common in plants and have certain nutritional and medicinal value.However,anthocyanins of Zijuan tea products were decreased in different degree during processing.It is important to explore the influence of the anthocyanin compositions in Zijuan tea under different processing.In this study,high-performance liquid chromatography-mass analysis(HPLC-MS)was used for qualitative and quantitative analysis of anthocyanin components in Zijuan tea under different processing.Furthermore,both metabolome and transcriptome were used to analyze the differential metabolites and differential genes of structural enzymes related to anthocyanin synthesis in purple leaves and green leaves of Zijuan.The main results were as follows:1.In this paper,both contents and species of anthocyanins were detected in 11 kinds of Zijuan tea and its related products using HPLC-MS.The results showed that Zijuan tea contained 6 anthocyanins,including cyanidin,pelargonidin,pelargoni din-3-O-glucoside,delphinin-3-O-galactoside and peonidin.Additionally,Delphinin-3-o-galactoside and cornflower were the main components in Zijuan tea.Under different processing processes,the species and contents of anthocyanins were reduced in a different degree,and the processes of fermentation and drying had the greatest influence on species and contents of anthocyanins.2.Furthermore,both metabolome and transcriptome were performed using purple leaves and green leaves of Zijuan.It showed that the metabolites and genes related to upstream of anthocyanin synthesis pathway were enriched in purple leaves.Through metabolic analysis,both the contents of cyanidins and cyanidin-3-O-rutin in purple leaves were significantly higher than that in green leaves.In addition,the contents of anthocyanin 3-O-glucoside and kathrinol 3-O-robinia glucoside in purple leaves were higher,which were related to the up-regulation of key genes of anthocyanin synthesis,including CsPAL,CsC4H,Cs4CL,CsCHS,CsCHI,CsANS,CsDFR and CsUFGT.It laid a foundation for promoting anthocyanin synthesis and laying a molecular foundation for improving the content of anthocyanin in Zijuan tea.
