| Arisaema amurense Maxim It is a perennial herb of Araceae and Araceae.Its tubers can be used as medicinal materials and have high medicinal value.A.amurense grows for a long time,resources are scarce,and the quality of different sources is not uniform.With the promotion of clinical medication in recent years,wild resources have been mined wantonly,and a large number of A.amurense have been consumed.In order to ensure the quality and market supply of A.amurense,and to improve its economic and medicinal value,this paper studied the yield and active component content of three-year-old A.amurense under different fertilization combinations,and optimized the ultrasonic-assisted extraction of total flavonoids from its tubers by response surface methodology.The purpose is to provide a basis for the development and utilization of A.amurense.The research conclusions are as follows:1.In the regression model between the ratio of nitrogen,phosphorus and potassium and the tuber fresh weight(yield)of A.amurense,it is found that the single effect of nitrogen,phosphorus and potassium on the yield of A.amurense is nitrogen>potassium>phosphorus.The fertilization combinations with the highest yield were N0P2K2and N2P0K2,and the fresh weight of tubers per plant reached 15.74g and 15.48g respectively.2.Through the analysis of variance after the optimization of response surface ultrasonic extraction process conditions,it can be seen that the influence of the three factors on the extraction results of total flavonoids is A solvent concentration(%)>B extraction time(min)>C material liquid ratio(g/ml).Response surface optimization showed that the optimum extraction conditions of ultrasonic assisted extraction of total flavonoids from the tubers of A.amurense were as follows:ethanol concentration 61%,extraction time 51 min and solid-liquid ratio 1:62(g/ml).3.Under the best extraction conditions of total flavonoids,the content of total flavonoids in the A.amurense of tubers of 10 different nitrogen,phosphorus and potassium treatments was determined.The results showed that the content of flavonoids in N2P0K treatment was the highest(2.33 mg·g-1),which was significantly different from other treatments.4.The contents of guanosine,adenosine and schaftoside in 10 kinds of A.amurense treated with different ratio of N,P and K were determined by HPLC.The highest content of guanosine was N3P3K1,reaching 313.53μg·g-1.In addition,the guanosine contents of N0P2K2 and N2P0K2 were also high,respectively 291.19μg·g-1、298.56μg·g-1,there was no significant difference with N3P3K1.There was no significant effect of different ratio of nitrogen,phosphorus and potassium on the contents of adenosine and schaftoside.5.The weight of three-year-old tubers of A.amurense and the content of flavonoids in tubers were comprehensively compared under different fertilizer treatments.The optimal fertilization scheme of nitrogen,phosphorus and potassium was selected,and the optimal treatment of N0P2K2(N 0g/m2、P2O5 8.94g/m2、K2O 10.73g/m2)and N2P0K2(N 13.42g/m2、P2O5 0g/m2、K2O 10.73g/m2)was determined. |
