Rape(B.napus)is the largest self-produced oil crop in China.The main producing area is the middle and lower reaches of the Yangtze River.It has the advantage of not competing for land with rice.However,the high water content of rice fields hinders the growth of rapeseed.In this study,the dry seeds of Brassica napus soaked in brassinolide were used as materials,and a new method to increase the yield of rapeseed was explored by using the existing seeds and field management.The main results are as follows:1.Transcriptome sequencing of the two processed materials and statistical analysis of the data through a variety of bioinformatics methods by Illumina high-throughput sequencing technology.The results are as follows: 466,286,03 and 467,844,66 clean reads were detected in the test group and the control group,screened out fatty acid synthesis-related differential genes(Bn FAT,Bn BCCP,Bn SAD)and photosynthesis-related differential genes(Bn Cab,Bn PSAG,Bn PSB28),and designed primers based on the above gene sequences using premer5.0 software,quantitative PCR was performed to verify,the expression of all 6 genes was up-regulated,the results was consistent with the sequencing results,which proved that the sequencing results were accurate.+2.Measure the physiological and biochemical indexes of the two groups of materials at different growth periods,use the fluorescence quantitative PCR technology to determine the expression of different genes related to fatty acid synthesis and photosynthesis of the two groups of materials,and analyze the correlation between the expression of the above-mentioned differential genes and the contents of physiological indexes.The results are as follows: Fatty acid synthesis-related differential genes Bn FAT and Bn BCCP are positively correlated with soluble sugar content,with correlation coefficient of 0.69;photosynthesis-related differential genes Bn Cab and Bn PSAG are also correlated with physiological and biochemical indicators,among which Bn Cab is associated with soluble protein content and SOD The activity is positively correlated with correlation coefficients of0.89 and 0.62.Bn PSAG and POD activity are positively correlated with a correlation coefficient of 0.6.The above results indicate that there is a close correlation between the differential gene expression and the content of physiological and biochemical indexes,and the later yield and quality of rapeseed can be predicted by monitoring the changes of these indexes.
