| Objective : Compound Rhubarb preparation is composed of rhubarb,Salvia Miltiorrhiza,Rhizoma Curcumae,radix scutellarlae and Radix eucommiae.It is used for treating deficiency and excess,inappetence and nausea caused by turbidity and toxin,and is the main manifestation of early stage and chronic kidney disease of kidney disease.In this study,the effect of compound rhubarb preparation on different types of rats with renal dysfunction was studied to provide experimental basis for veterinary clinical application.Methods:1.Effects of Compound Rhubarb preparation on unilateral ureteral obstruction and partial nephrectomy induced renal fibrosis model in rats: 140 male SD rats were randomly divided into 12 groups,20 rats in blank control group,20 rats in sham operation group,10 rats in unilateral ureteral obstruction model group,10 rats in right kidney model group by ligation of surgical line,and 10 rats in Niaoduqing granule(7.2 g / kg weight)The positive control group was treated with Niaoduqing granules(7.2 g / kg weight)for unilateral ureteral obstruction,the positive control group was treated with surgical ligation and resection of the whole right kidney,and the experimental groups were treated with Compound Rhubarb preparation(1.94 g,3.87 g,7.74 g/ kg weight)for unilateral ureteral obstruction,Thirty right kidneys were ligated and excised with surgical thread.Three experimental groups were treated with Compound Rhubarb preparation: low,medium andhigh(1.94 g,3.87 g,7.74 g/ kg weight)Three experimental groups.After 28 days of medication test,blood was collected to detect serum creatinine(crea)and urea nitrogen(urea),urine volume was collected and measured,24-hour urinary protein(pro)and creatinine(crea)were detected,and glomerular filtration rate(e-gfr)was calculated.Then,the obstructed kidney and the partial nephrectomy group were removed and stored in neutral formalin.h-E staining and Masson staining were performed.histological sections were observed,described and micrographed.2.Effect of Compound Rhubarb preparation on rat model of cold coagulation and blood stasis: 70 SD rats,half male andhalf female,were randomly divided into 7 groups according to body weight: blank control group,model control group,Niaoduqing granule group(7.2 g / kg),aspirin enteric coated tablets group(54 mg / kg),low,medium andhigh dose compound rhubarb preparation groups(1.94,3.87,7.74 mg / kg)The rats in each group were given the corresponding dose of liquid medicine by gavage,once a day for 7 consecutive days.On the sixth day of administration,0.8 mg / kg epinephrinehydrochloride injection was subcutaneously injected 1h after administration.After 2hours,the animals were immersed in ice water at 4 ℃ for 5 min,and then taken out.After 2hours,the same dose of epinephrine was injected again.After1 hour of administration on the next day,the blood was anesthetized and thehemorheological indexes were detected.Result:1.Effect on serum crea content: compared with the blank control group,the serum crea levels of model control group,low,medium andhigh dose rhubarb preparation groups and Niaoduqing granule group were significantly increased before administration(P < 0.01),and there was no significant difference in other groups.Compared with the blank control group,the serum urea of model control group,low,medium andhigh dose rhubarb preparation groups and Niaoduqing granule group were significantly increased before administration(P < 0.01),and there was no significant difference in other groups.Compared with the blank control group,the content of crea in urine of the model control group was significantly increased after 4 weeks of Administration(P < 0.05),and there was no significant difference among other groups.Compared with the blank control group,the e-gfr of the model control group and the low-dose rhubarb compound preparation group were significantly decreased after 4weeks of Administration(P < 0.01),and there was no significant difference in the egfr of the sham operation group,the medium andhigh-dose rhubarb compound preparation group and the Niaoduqing granule group after 4 weeks of Administration(P > 0.05);compared with the model control group,the e-gfr of the medium andhighdose rhubarb compound preparation group and the Niaoduqing granule group were significantly decreased after 4 weeks of administration It was significantly increased(P< 0.01).Compared with the blank control group,the kidney organ coefficient of lowdose rhubarb compound preparation group was significantly increased(P < 0.05),and that of model control group was significantly increased(P < 0.01).There was no significant difference in kidney organ coefficient among sham operation group,medium dose rhubarb compound preparation group,high-dose rhubarb compound preparation group and Niaoduqing granule group(P > 0.05).2.The results of microscopic observation showed that: in the blank control group and sham operation group,the structure of renal cortex and medulla was clear,the morphological structure of nephron was normal,and there was no inflammatory cell infiltration in the interstitial,in the model control group,there was degeneration and necrosis of renal tubular epithelium,lumen expansion,proliferation of interstitial fibrous connective tissue,and inflammatory cell infiltration;compared with the model control group,the degree of pathological changes in each drug group was significantlyhigher According to the severity of the disease,the order from severe to mild was compound rhubarb preparation low dose group,compound rhubarb preparation medium dose group,Niaoduqing granule group,compound rhubarb preparationhigh dose group.The results of Masson staining showed that the positive expression of blank control group and sham operation group was less,while the positive expression of model control group was the most.Compared with model control group,the positive expression of each administration group was lighter.According to the positive expression rate,from more to less,the order was compound rhubarb preparation low dose group,compound rhubarb preparation medium dose group,Niaoduqing granule group,compound rhubarb preparationhigh dose group The results are in agreement with each other.3.Effect of Compound Rhubarb preparation on rat model of cold coagulation and blood stasis: compared with the blank control group,the whole blood viscosity of low,medium,high shear and plasma viscosity of rats in the model control group of cold coagulation and blood stasis syndrome were significantly increased(P < 0.01);compared with the model control group,the whole blood viscosity of low,medium,high shear and plasma viscosity of rats in the medium dosage group of Compound Rhubarb preparation were significantly decreased(P < 0.05)The low,medium andhigh shear values of whole blood viscosity and plasma viscosity were significantly decreased inhigh-dose rhubarb preparation group,Niaoduqing granule group and aspirin enteric coated tablet group(P < 0.01).Conclusion:Compound Rhubarb preparationhas the effects of purging fu-organs and reducing turbidity,invigorating spleen and removing dampness,promoting blood circulation and removing blood stasis in rats,ithas obvious therapeutic effect on the model of renal fibrosis caused by unilateral ureteral obstruction,the model of renal fibrosis caused by partial nephrectomy and the model of cold coagulation and blood stasis. |
