| Purpose: In this experiment,adenine was used to replicate the rat model of Chronic renal failure(CRF),and Lishhen formula was applied for drug intervention on the CRF rat model.The therapeutic effect of Lishhen formula on the CRF rat model was observed from various aspects,and the influence of Lishhen formula on CRF rats was discussed.To provide prescription treatment ideas and scientific theoretical basis for the treatment of chronic renal failure with traditional Chinese medicine.Methods: 60 healthy male SD rats were randomly divided into six groups: blank control group,model group,positive control group(3.6g/mL),Lishhen formula low-dose group(2.4g/mL),Lishhen formula medium-dose group(4.8g/mL)and Lishhen formula high-dose group(9.6g/mL).After 28 days of intragastric administration of 2.5% adenine suspension,Serum creatinine(Scr),blood urea nitrogen(BUN)levels,clinical symptoms,renal index and pathological structure were detected to judge the success of modeling.After 21 days of treatment,clinical symptoms,renal index and renal pathological changes(HE staining and MASSON staining)were observed,Scr,BUN,Malondialdehyd(MDA)and Super Oxide Dismutase were detected.SOD),Interleukin-1(IL-1β),Interleukin-1(IL-6),Tumor Necrosis Factor-α(TNF-α),Laminin,LN),Fibronectin(FN),CollagenⅣ(ColⅣ)levels and Transforming growth factor-β of kidney tissue were detected by transforming growth Factor-βimmunohistochemistry.TGF-β),Phosphoinositide 3-kinase(PI3k),Phosphoinositol 3 kinase,Phosphorylated protein kinase B(p-Akt)protein expression.Results: Compared with before treatment,the clinical symptoms of rats in all treatment groups were improved,and the effect of Lishhen formula high dose group was significant(p<0.01).After treatment,the contents of serum Scr and BUN in the treatment group were significantly lower than those in the model group(p>0.01),and the kidney index was decreased.Pathological results showed that the damage degree of kidney tissue in treatment groups was significantly lower than that in model group.Compared with model group,the contents of MDA and SOD were decreased,the contents of IL-1β,TNF-α and IL-6 were significantly lower than those of model group(p>0.01),and the contents of LN,ColⅣ and FN were decreased(p>0.05,p>0.01).The protein expressions of TGF-β,p-PI3 K,PI3k and P-Akt in the treatment group were significantly lower than those in the model group(p>0.01). |
