Preventive Effects Of Taurine On Hyperuricemia Nephropathy In Goslings

Abstract/Summary:
Hyperuricemia nephropathy is a common metabolic disease in goslings and caused by high level of uricemia and accompanied by kidney damage.Our previous study finds that taurine can reduce the level of hyperuricemia in rats and reverse kidney damage.However,the preventive effect and regulatory mechanism of taurine on hyperuricemia nephropathy in goslings are not clear and need further study.Sixty 10-day-old goslings are randomly divided into 4 groups with 15 goslings in each group.Control group(C)is fed full price goose feed.Hyperuricemia nephropathy model group(M)is fed a high-protein and high-calcium diet,taurine prevention group(MT)is fed a high-protein and high-calcium diet +0.1% taurine,and taurine control group(T)is fed a full-price goose diet +0.1% taurine.The other feeding conditions are normal,and the geese are fed for 21 days.Serum,liver and kidney are collected for follow-up tests.The methods of IHC,q RT-PCR and ELISA are used to study the effect of taurine on the prevention of hyperuricemia nephropathy in goslinger.The test results show that:1.Compared with group C,liver index and kidney index are increased in group M,liver and kidney function are impaired,and kidney tissue is damaged.However,there is no significant difference in renal index and liver and kidney function between MT group and control group(P>0.05).In group M,uric acid production increased,reabsorption ability increased,excretion ability decreased.The activities of XOD and ADA in serum and liver and the mRNA expression levels of XOD and ADA in liver of M group are significantly increased(P<0.05),and the mRNA expression levels of GLUT9 and URAT1 related transporters of renal uric acid reabsorption are significantly increased(P<0.05).The mRNA expression levels of OAT1 and OAT3 related transporters in uric acid excretion are significantly decreased(P<0.05).While MT group can reverse the above changes,taurine can prevent kidney damage and reverse kidney damage caused by high uric acid.2.Compared with group C,inflammation level in group M is increased,the contents of IL-1β,IL-6 and TNF-α in serum,TNF-α,IL-1β and MCP-1 in kidney are significantly increased(P<0.05),The mRNA expression of TLR4,My D88,NF-κB,NLRP3,ASC in kidney are significantly increased(P<0.05).The mRNA expression of Caspase-1 is significantly increased(P<0.05).The inflammation level of MT group is decreased,and taurine can alleviate the inflammation and injury of hyperuricemia nephropathy gosling.3.Compared with group C,the antioxidant level of group M is decreased,SOD content in serum and kidney of group M is significantly decreased(P<0.05),MDA content is significantly increased(P<0.05),The mRNA expression levels of Nrf2,HO-1,γ-GCS and NQO1 in kidney are significantly increased(P<0.05).The antioxidant level of MT group increased,and taurine can reverse the antioxidant damage.4.Compared with group C,renal tubulointerstitial fibrosis occurred in group M,the serum TGF-β content in group M is significantly increased(P<0.05),and the mRNA expressions of α-SMA,COL1A1,FN,β-catenin and Wnt in group M are significantly increased(P<0.05).The expression of α-SMA and COL1A1 in renal tubule interstitium is significantly increased(P<0.05).The level of tubulointerstitial fibrosis decreased in MT group,and taurine can inhibit the occurrence of tubulointerstitial fibrosis.The results of this study showed that taurine can effectively reduce the level of uric acid in goslings with hyperuricemia nephropathy,and prevent hyperuricemia nephropathy by alleviating renal inflammation,reversing antioxidant damage and inhibiting the occurrence of renal tubulointerstitial fibrosis.
