Oats are an important grain crop in China and a dominant crop in Inner Mongolia.Oats are rich in β-glucan and have high nutritional value.Whole genome resequencing technology can obtain a large number of SNPs for molecular genetic markers,providing an important reference for crop genetic breeding and variety selection.In this study,130 oat germplasm seeds were evaluated for β-glucan content,and the whole-genome resequencing analysis was carried out to select BY18 and DY5 with extreme differences in β-glucan content,to obtain SNPs related to β-glucan content,andq RT-PCR was performed to verify the SNPs related toβ-glucan content in oat seeds,and to provide a basis for the development of molecular markers and identification of β-glucan in oats.The main results of the study are as follows:1.Based on β-By cluster analysis of glucan content,the tested germplasms were divided into three categories.The first category contained 76 oat germplasms,β-Dextran content was medium,with an average of 3.90%;There are 5 oat Germplasms in the second category,β-Dextran content was the highest,with an average of 5.11%;The third category includes 49 oat germplasms,β-Dextran content was the lowest,with an average of 2.62%.2.The results of high-throughput sequencing showed that the original data of Diyan 5 was49.5g,and that of bayou 18 was 50.7g.After filtering,the effective data of Diyan 5 was49.0g,and that of bayou 18 was 50.1g.3.Comparing the whole genome re sequencing data with oat reference genome data,4146177 SNP sites were detected in Diyan 5 and 6176966 SNP sites were detected in bayou18.Twenty five SNP loci were screened from the functional annotation of Diyan No.5 and Ba you No.18 genes,which fell on eight genes: pepsico1_Contig37478 、 Pepsico1_Contig39237、Pepsico1_Contig37825、Pepsico1_Contig15205、Pepsico2_Contig10922、Pepsico2_Contig18947、Pepsico2_Contig13535、Pepsico2_Contig16295。 All 8 genes are related to control β-The gene family CSL related to glucan synthesis pathway has high homology.4.The results of fluorescent quantitative PCR showed that the 8 genes were expressed at grain filling stage,β-Dextran related SNP sites are real.
