| Populus spp.has the advantages of strong adaptability and rapid growth,and is regarded as a model plant in the cultivation and breeding of forest trees.In recent years,China has introduced,cultivated and promoted a large number of new poplar varieties and excellent clones,but many varieties have a series of problems,such as regional adaptation is not clear,the morphological traits are not obvious,affected by the environment,the name is chaotic,etc,which have had an adverse impact on promotion and application of new poplar varieties.In this study,26 superior poplar clones(variety)were cultivated in recent years,and regional comparative experimental forests were established in Luannan,Yanshan and Fucheng of Hebei Province.The growth performance and adaptability of different clones in different regions of Hebei Province were compared and analyzed in order to initially screen out suitable clones.At the same time,94 superior poplar clones and varieties suitable for growth in Northeast China were collected,and SSR molecular marker technology was used to construct DNA fingerprints of the varieties,and molecular identification and genetic diversity analysis were carried out to provide protection for the screening and popularization of new varieties.The main findings are as follows:1.The survey of the two-year-old experimental forests in Luannan,Yanshan and Fucheng showed that there were significant differences in DBH,tree height and survival rate among the clonal lines in the three experimental forests,and the performance of clones in different test sites is different.The clones with excellent growth performance in the Fucheng test forest were 108,2001 and 111;the clones with good growth performance in the Luannan test forest were 107,L2 and BF-1;the clones with good growth in the Yanshan test forest were BF-2,L2 and 111.2.The combined analysis of variance of six poplar clones at three locations showed significant differences in DBH and tree height in the interaction between clones and locations.The stability analysis of the variety showed that 108 poplar had good growth performance in the Fucheng;BF-2 poplar was excellent in the Yanshan;L2 poplar had wide adaptability and performed well in all three experimental sites and could be promoted.3.Two clusters of the clones were tested by 14 pairs of SSR primers.Cluster analysis of 94 poplar clones can distinguish all clones.All poplar clones have specific bands and rich genetic diversity.Further cluster analysis of 54 poplar clones with clear origins can distinguish poplar clones according to their relative relationship according to the three major groups of Aigeiros,Leuce and Tacamahaca,which consistent with the origin of the clones and prove that the primers used are representative4.94 pairs of poplar clones were constructed using 14 pairs of SSR primer combinations with good amplification and reproducibility.The fingerprint is composed of the fingerprint code consisting of the primer name and the size of the band,and is compiled into the database for identification of new varieties of poplar. |
