Beef as an essential food in people’s daily lives,has a high nutritional and economic value.Prohibited veterinary drugs are a type of drugs that are explicitly prohibited from being used in animal based food in national regulations.These drugs are highly toxic and harmful,and their carcinogenic and mutagenic effects pose great harm to human health and the ecological environment.Therefore,the residual issue of prohibited veterinary drugs in animal based food has always been a focus of attention at home and abroad.Liquid chromatography tandem mass spectrometry(LC-MS)is a commonly used technology for modern detection of veterinary drug residues in food.High resolution time-of-flight mass spectrometry has high resolution and excellent accuracy.This study combines liquid chromatography high-resolution time-of-flight mass spectrometry to establish an efficient and rapid high-throughput screening and detection technology for prohibited veterinary drugs in beef,and applies it to the detection of prohibited veterinary drug components in commercial beef,in order to provide theoretical basis for regulatory authorities to detect prohibited veterinary drugs in beef.Using liquid chromatography high-resolution time-of-flight mass spectrometry to screen 17 prohibited veterinary drugs in beef,the samples were extracted with 0.2%formic acid(80:20)acetonitrile aqueous solution,KNORTH~?Clean up LPAS purification column purification.Using the Phenomenex Kinetex F5 chromatographic column,0.1%formic acid water and 0.1%formic acid acetonitrile were used as the positive ion scanning mobile phase,and water and acetonitrile were used as the negative ion scanning mobile phase for gradient elution.The flow rate was 0.3m L/min,and the column temperature was 40℃.The mass spectrum was scanned using an electrospray ion source,which was divided into positive ion and negative ion scanning modes.The detection limit(LOD)and quantification limit(LOQ)of 17prohibited veterinary drugs in beef are 0.04-0.28μg/kg and 0.12-0.95μg/kg.The recovery rates of 17 prohibited veterinary drugs in beef were determined,ranging from 75.7%to 102.0%.17 prohibited veterinary drugs in 5-200 The linear relationship is good within the range ofμg/m L,with R~2 greater than 0.98.Methodological examination was conducted on the established method,and the recovery rate,detection limit,quantification limit,etc.obtained met the requirements of national standards.100 samples of commercially available beef were collected for actual sample testing.Among the 17 prohibited veterinary drugs,only clenbuterol was detected,while the rest were not detected.The high-throughput screening technology established by our research institute is suitable for the detection of 17 prohibited veterinary drugs in beef.This method has the advantages of fast and simple detection,and can effectively detect prohibited veterinary drug components in beef.It has high accuracy and reliability,making up for the shortcomings of previous detection methods such as single drug types and complex sample pre-treatment.
