Cornus officinalis Sieb.et Zucc.is a deciduous tree or shrub species,belonging to the family Cornaceae and the genus Cornus L.It is also an important medicinal,edible and ornamental plant.The genetic diversity and genetic structure of germplasm resources of C.officinalis in Anhui Province were investigated based on ISSR markers.Results of the present research are as follows:1.The genetic diversity of germplasm resources of C.officinalis in Anhui Province was investigated,Results revealed a medium level of the resources.Thirteen polymorphic effective ISSR primers were selected.The genetic diversity was explored at 349polymorphic loci with 204 samples from 10 populations of C.officinalis,using 13 selected ISSR primers by agarose gel electrophoresis.At the species level,the results showed the percentage of polymorphic loci PPLs=100%,the number of effective alleles Nes=1.437,Nei’s gene diversity(expected heterozygosity)Hes=0.258,and the Shannon’s information index Is=0.429.At the population level,the results showed the percentage of polymorphic loci PPLp=70.46%,the number of effective alleles Nep=1.343,Nei’s gene diversity Hep=0.210,Shannon’s information index Ip=0.325.2.The genetic structure of germplasm resources of C.officinalis in Anhui Province was explored.Results indicated a medium level of genetic differentiation among populations.The genetic variation was mainly found within populations,There was no significant genetic difference between Dabie mountain area and southern Anhui mountain area.The coefficient of genetic differentiation among populations(Gst)was 0.179.Analysis of molecular variance(AMOVA)showed the genetic variation among and within populations accounted for 23.05%and 76.95%of the total genetic variation,respectively.The genetic distances between populations ranged from 0.029 to 0.068.Mantel test also showed that there was no significant correlation between geographical distance and genetic distance.The strategies of conservation and utilization of genetic diversity of C.officinalis in Anhui Province were suggested based on the above resultsResults of the present research will provide theoretical basis and practical guidance for conservation and utilization of germplasm resources of C.officinalis,and sustainable development of industry of the species in Anhui Province.
