| Peanut sprouts,also known as long-life sprouts,are a kind of therapeutic food produced by peanut kernels,which is favored by many people nowadays.In this study,181 peanut germplasm resources were selected for the production of peanut sprouts by measuring their shoot length,shoot yield and weight-gain rate,followed by measuring the physiological parameters of these germplasm resources and finally transcriptome sequencing between three germplasm resources,control and under UV treatments,differential developmental stages were conducted.Results were obtained as following:(1)Germination experiments were carried out by hydroponics,and the shoot length,shoot yield and weight-gain rate were measured on the 7th day to screen for the germplasm resources suitable for peanut sprout production,and the results showed that JND44,JNP310,ZY131 and HY16 could be used for peanut sprout production with shoot length longer than 10 cm,shoot yield greater than 95%and weight gain rate more than 330%.(2)The physiological indexes related to edible quality were determined for sprouts of HY 16,JNP310,ZY131.The results showed that the ash contents of ZY131 and HY 16 were significantly lower than that of JNP310,the protein content of ZY131 was significantly higher than that of JNP310,the fat content of HY 16 was significantly lower than that of ZY131 and JNP310,the crude fiber content of JNP310 was significantly lower than those of HY 16 and ZY131,and the soluble sugar content of HY 16 was significantly lower than those of JNP310.Totally,the results indicated that edible quality of HY16 were better than those produced of JNP310 and ZY131.(3)Expression analysis of differential genes between groups clarified the gene expression change patterns of peanut sprouts at different developmental stages,different germplasm resources and UV treatments.The growth and developmental stages of peanut sprouts are influenced by circadian rhythms and accompanied by the accumulation of secondary metabolites such as resveratrol.The expression of secondary metabolism-related genes in peanut sprouts were induced by UV treatment,and resveratrol synthesis genes were significantly induced,with the highest expression of STS genes at 600 s under UV treatment.Three STS genes were expressed in all three germplasm sources of peanut sprouts,and the STS expression of HY16 was higher than those of JNP310 and ZY131. |
