As a major agricultural province in the northwest region of China,Xinjiang is one of the regions that have suffered the most severe damage from invasive organisms in agriculture and forestry.After the introduction of Liriomyza sativae(Blanchard),Liriomyza huidobrensis(Blanchard),Phthorimaea absoluta and Frankliniella occidentalis into Xinjiang,they pose a potential threat to Xinjiang’s agricultural production through the use of facility agriculture as a bridge.This article conducts risk analysis on the Liriomyza sativae Blanchard,Liriomyza huidobrensis(Blanchard),Phthorimaea absoluta and Frankliniella occidentalis,predicts their suitable distribution range,and uses loop mediated isothormal amplification(LAMP)technology to develop detection techniques for invasive organisms such as Phthorimaea absoluta and Ambrosia(Ambrosia trifida L.and Ambrosia artemisiiiflia L.).The main conclusions are as follows:1.Based on the current climate conditions,the MaxEnt ecological niche model was used to predict the distribution of potential suitable habitats of four small insects in Xinjiang.Thc results showed that the Phthorimaea absoluta high-suitability areas and suitability areas accounted for about 28.34%of the total area in xinjiang,mainly distributed in the north with,turpan city,northwest once.during north-central course,shihezi,western region,central aksu area,the parts of kashi,hotan region.The high suitability areas and suitability areas of The Liriomyza sativae Blanchard accounted for 38.58%of the total area of Xinjiang,and the potential suitability areas were mainly concentrated in Changji Prefecture and Turpan Prefecture in northern Xinjiang,Kashgar Prefecture and Bazhou Prefecture in southern Xinjiang.The high suitability areas and suitability areas of the Liriomyza huidobrensis(Blanchard)account for 20.39%of the total area of Xinjiang,and the potential suitability areas include part of Hami City,Changji Prefecture,northern Turpan City,Karamay District,part of Bazhou,central Aksu Prefecture,part of Kezhou Prefecture,most of Kashgar Prefecture and central Hotan Prefecture.The highly suitable areas and suitable areas of Frankliniella occidentalis accounted for 43.56%of the total area of Xinjiang,and the potential suitable areas in Xinjiang were mainly concentrated in Changji Prefecture,Hami City and Turpan City in northern Xinjiang,and Kashgar Prefecture and Bazhou in southern Xinjiang.The Altai Mountain range,Tianshan Mountain range and Kunlun Mountain range,as well as the Zhuanger Basin and Tarim Basin desert areas are not suitable for the four species of insects.According to different levels of invasion risk in different areas,more efficient and targeted monitoring and prevention can be carried out to save the waste of human and material resources.2.Using COI gene of Phthorimaea absoluta as target,LAMP detection system of Phthorimaea absoluta was established.Nucleic acid amplification reaction was performed for 60min at 65℃ isothermal condition.After amplification,SYBR GrecnI dye was added into the system.After the reaction,the color change was directly observed by naked eye to determine the result(positive was fluorescent green,negative was orange).The minimum detection limit of DNA by this method is 2.08-4ng·μL-1.This method provides a quick and accurate technique for the field detection of Phthorimaea absoluta moth.3.LAMP technology was developed for Ambrosia trifida L.and Ambrosia artemisiifolia L.,taking SYBR Green I as indicator,to achieve rapid,accurate and efficient field identification of Ambrosia trifida L and Ambrosia artemisiifolia L.Specificity verification results showed that LAMP products of the tested weeds were all positive(white precipitates),while LAMP products of the other two weeds in the control were negative(no white prccipitatcs).The sensitivity test results showed that the minimum detection limit of DNA of the system was 10-10ng·μL-1,which was higher than that of conventional PCR.The LAMP detection system established in this study can be effectively applied to the rapid detection of Ambrosia trifida L and Ambrosia artemisiifolia L samples,providing technical support for its rapid and efficient identification.
