| Toxicodendron vernicifluum is a kind of economic tree species with a long history.Its raw lacquer,lacquer oil and lacquer wax are important raw materials for many industries in today’s society.However,many wild lacquer tree plus tree resources have not been well protected and developed,and the problems of root cutting and tissue culture and rapid propagation technology need to be further improved and improved,which seriously restrict the development of lacquer tree industry.In this study,the wild lacquer tree superior tree resources and lacquer tree varieties(Da Hong Pao,Jin Gangda Muqi and Gao Ba Chi,etc.)in Pingli County and Langao County of Ankang City,Shaanxi Province were used as research materials to explore the survival rate and genotype differences of root cutting propagation of different wild lacquer tree superior tree resources,and to analyze the differences in photosynthesis,water use efficiency and growth traits of different types of lacquer trees.The differences of photosynthesis,water use efficiency and growth traits of lacquer tree under different hormone treatments were analyzed by using lacquer tree’Gaobachi’as material.On this basis,the best disinfection method of explants,proliferation and adventitious root induction culture method were studied.Through this study,19 wild lacquer tree plus trees were collected and asexually propagated and preserved by root cutting.The best hormone combination for root cutting of lacquer tree was screened out by using lacquer tree’Gaobachi’as material,and the cutting propagation system of lacquer tree root was optimized.The tissue culture and rapid propagation of lacquer tree were carried out with the same clone of wild lacquer tree No.69 and lacquer tree’Gaobachi’.The main conclusions are as follows:(1)Root cutting propagation was carried out on the selected wild lacquer tree.The highest survival rate of cutting was 60%,the lowest was 10%,and the average was 37.4%,19 wild lacquer tree plus tree resources were preserved.(2)Using lacquer tree improved variety’Gaobachi’as material.The application of 20 mg/L GA3+200 mg/L ABT could improve the survival rate of lacquer tree root cutting,from 60%to 76%.It was found that 100 mg/L ABT solution could improve the photosynthesis of lacquer tree seedlings.Applying 20 mg/L GA3could increase the stomatal conductance Gs and transpiration rate Tr of lacquer tree seedlings.(3)The best disinfection method of lacquer tree explants was determined by using wild lacquer tree and the same clone of lacquer tree variety’Gaobachi’was used as material.The young stem segments of lacquer tree were immersed in 75%anhydrous ethanol solution for 30 s,immersed in 2.0%bromogeramine(benzalkonium bromide)for 1.5 min,and immersed in 0.1%mercuric chloride solution for 5 min.It was confirmed that MS+2.0 mg/L 6-BA+0.5 mg/L NAA+0.8g/L AC was suitable for axillary bud germination of lacquer tree.MS+0.5 mg/L ZT+1.0 mg/L 6-BA+0.1 mg/L TDZ was suitable for the proliferation of lacquer tree seedlings.1/2MS+1.0 mg/L GA3+0.5 mg/L NAA was suitable for the induction of adventitious roots.In summary,this study preserved and expanded the excellent tree resources of wild lacquer tree,optimized the root cutting propagation system of lacquer tree,improved the survival rate of root cutting of improved varieties of lacquer tree,and realized the preservation and propagation of wild lacquer tree and improved varieties of lacquer tree through tissue culture and rapid propagation technology,which provided important technical support for the development of lacquer tree industry. |
