With the improvement of people’s living standards around the world,the incidence rate and mortality of ulcerative colitis in people and pets have also increased,which has been listed as an important public health issue by the World Health Organization.In the past,the treatment methods for this disease mainly focused on the following: drug treatment,surgical resection,nutritional agents and biological agents.However,due to the uncertainty of the cause of ulcerative colitis and the complexity and variability of symptoms,the above treatment methods will have a certain negative impact on patients,and may cause other complications in serious cases.Therefore,more and more people begin to explore new methods of treating ulcerative colitis with traditional Chinese medicine preparations.Traditional Chinese medicine preparation has unique curative effect in treating diseases with complex etiology.Its side effects are controllable and its curative effect has been verified for thousands of years.The combination of multiple herbs can reduce the toxicity of drugs and enhance the therapeutic effect of the monarch medicine on diseases.Based on the theory of Chinese veterinary medicine,this experiment studied the therapeutic effect of various combinations of medicinal materials on ulcerative colitis model mice,and tried to screen out the effective combination of drugs to make contributions to the research of special prescription for the treatment of ulcerative colitis.Experiment 1: According to the incompatibility of drugs,draw up three traditional Chinese medicine prescriptions.In order to explore the therapeutic effect of Qingchang Tiaoxue recipe,Jianpi Qushi recipe,Tiaogan Lipi recipe on ulcerative colitis induced by dextran sodium sulfate in mice.In the experiment,66 female C57BL/6 mice(6~8 weeks old,18~20 g)were randomly divided into 11 groups,6 in each group.Blank control group(Group K),model group(Group S),high dose Qingchang group(Group A),medium dose Qingchang group(Group B),low dose Qingchang group(Group C),high dose Jianpi group(Group D),medium dose Jianpi group(Group E),low dose Jianpi group(Group F),high dose Tiaogan group(Group G),medium dose Tiaogan group(Group H),and low dose Tiaogan group(Group I).In the first week,each group was fed adaptively.In the second week,in addition to the blank control group,each group drank 3% dextran sodium sulfate aqueous solution freely for 7 days to establish an acute ulcerative colitis model.In the third week,the normal drinking water was replaced.The blank control group and the model group were given physiological saline by gavage.The drug groups were given different concentrations by gavage.The clinical manifestations of the mice were observed and recorded.The whole blood serum was collected and the serum immunoglobulin G(Ig G)was detected,The mass concentration changes of immunoglobulin A(Ig A),interleukin-8(IL-8),interleukin-10(IL-10),diamine oxidase(DAO),D-lactic acid,and the pathological sections of the mouse colon were made.The results showed that the mass concentration of D-lactic acid,interleukin-8(IL-8),immunoglobulin G(Ig G),immunoglobulin A(Ig A),diamine oxidase(DAO)in the model group was significantly higher than that in the blank control group(P<0.01),and the mass concentration of interleukin-10(IL-10)was significantly lower(P <0.01).After administration,the mice in 9 groups of Qingchang Tiaoxue,Jianpi Qushi and Tiaogan Lipi had different degrees of inflammation relief,The mass concentrations of D-lactic acid,IL-8,Ig G,Ig A and DAO decreased to varying degrees,while the mass concentrations of IL-10 increased.It shows that the prescription of clearing the intestines and regulating blood,invigorating the spleen and removing dampness,and regulating the liver and regulating the spleen have good therapeutic effects on DSS-induced ulcerative colitis.Experiment 2: To explore the effects of the proposed three traditional Chinese medicine formulations on the intestinal microbial community in mice with ulcerative colitis.The occurrence of ulcerative colitis is often accompanied by changes in the abundance and structure of the intestinal flora in the colon.Therefore,16 Sr RNA amplification sequencing technology was used to analyze the distribution of the flora in the colon contents of mice.The results showed that after the intervention of three traditional Chinese medicine formulations,the structure of the colonic flora in ulcerative colitis model mice was restored,which could increase the abundance of some beneficial bacteria such as Patescibacillus(P <0.05),Muribacoccaceae(P<0.05),Lactobacillus(P <0.05),Prevotellaceae UCG001(P <0.05),and reduce the abundance of some harmful bacteria such as Verrucomomicrobiota(P <0.05),Desulfobacterota(P <0.05)Abundance of Lachnospiraceae NK4A136 group(P <0.05),Campilobacterota(P <0.05),Actinobaciota(P <0.05),Bacteroidaceae(P <0.05),Tannerella laceae(P <0.05).
