As lactobacillus and Bifidobacterium andLactobaccillus rhamnosum typical intestinal probiotics, animal gastrointestinal tractenvironment, not only can tolerance to colonize the human intestinal tract regulatingintestinal flora, and it also has a prevention and treatment of diarrhea, eliminate toxin,enhance immunity, etc features; But its nutrition is a tall order, and only absorb doses in1x106cfu/mL or more, can give full play to the function of live. Nine common to spleenand stomach of Chinese herbal medicine as raw materials, Bifidobacterium, lactobacillusrhamnose as test strains, screening for probiotics in vitro growth of Chinese herbalmedicine, further by TNBS type ulcerative colitis rats model, to give better enrichmentextract of Chinese herbal medicine, observe the recovery of ulcerative colitis rats andchanges in intestinal flora.This paper first research divine comedy by turbidimetric method, YiMi, red ginseng,licorice root, Chinese yam, amomum fruit, atractylodes, poria cocos, nine kinds of spleenand stomach of Chinese herbal medicine combination of Bifidobacterium and lactobacillusrhamnose in vitro growth and the effect of three kinds of enrichment of Chinese herbalmedicine with better effect in vitro of probiotics, the divine comedy, YiMi, red ginseng,respectively, and the basal medium is extremely significant difference (p <0.01), and thenin at live bacteria counting method for the three kinds of traditional Chinese medicine(TCM) with6.25%,12.5%,25%,50%concentration of four traditional Chinese medicineculture medium with live bacteria, results showed that the concentration of25%of divinecomedy culture medium for in vitro growth of probiotics has the largest enrichment effect,and the basal medium difference is extremely significant difference (p <0.01). In vivo tests for ulcerative colitis rats as experimental model, were divided intocontrol group and the divine comedy, red ginseng, YiMi group three group, each group ofeight, only by TNBS/ethanol after the success of the method to build model, experimentalgroup according to the4g/kg dose lavage regularly every day to give placed at roomtemperature of the traditional Chinese medicine extract, model control group given dosesof distilled water, such as once a day, and test a total of14days. Daily observation recordsrats weight loss situation and stool properties and defecate occult blood, stool, DAI rating,sterile collecting dung, put to death before death in rats after the trial period, the colonpathological slice observation, determination of malondialdehyde in the colon andmyeloperoxidase levels, the Real-time PCR methods to analyze UC rat intestinal floradistribution. Control group ratscompared with the experimental group, showed the spleen, thymus atrophy, intestinalweight index increased. Pathological observation found that in the control group theexperimental group, the mucosa and submucosa has a large number of inflammatory cellsand a lot of hyperemia and edema, focal lymphocytic infiltration and muscular celldeformation. Intestinal flora in the experimental group compared with control group, thenumber of e. coli showed a trend of decrease, lactobacillus number is on the rise,bifidobacterium has no obvious trend. Showed that the divine comedy, red ginseng, YiMiextract for the prevention and treatment of UC rats has a certain role.So25%of the divine comedy medium as the foundation, and other enrichment effectis good, red ginseng, YiMi to best select enrichment are optimized by the orthogonalexperiment of traditional Chinese medicine compound medium, Bifidobacterium animalsenrichment medium optimal proportion for the divine comedy25%, YiMi5%, red ginseng,10%,37℃cultivate16h living bacterium number up to2.26x1011CFU/mL, the basalmedium and cMRS was medium increased by525,52times; Bifidobacterium longenrichment medium optimal proportion for the divine comedy25%, YiMi5%, red ginseng,5%,37℃for16h, the maximum1.64x1011cfu/mL, respectively raised the basicculture medium and cMRS medium than control,886,20times; Lactobaccillusrhamnosum bacteria culture medium formula for increasing the divine comedy, YiMi5%,red ginseng,10%25%,37℃when train13h number of living bacterium of3.52x1011CFU/mL, the basal medium and cMRS was medium increased by550,43times.This test for the relationship between the spleen and stomach of Chinese herbalmedicine and probiotics was system studied, for the development of new Chinese herbalmedicine health care products and probiotic products to provide a theoretical basis. |
