It is generally believed that the liver is an organ for lipid metabolism,and the liver and adipose tissue are the main targets for fat synthesis and storage,but lipids are mainly digested and absorbed in the intestine.We found that dietary supplementation of 0.025g·kg-1 RES had a significant effect on the expression of related genes in the fat synthesis step of tilapia,and changed the structure of tilapia intestinal flora and the concentration of metabolites of the flora.Taking red tilapia as the research object,construct 8%normal fat group and 10%high fat group,10%high fat group+0.025 g·kg-1 RES and 10%high fat group+0.025 g·kg-1RES+5m M EX527(inhibitor)four experimental groups:use HE staining to observe the morphological changes of red tilapia midgut,ELISA to detect midgut immune factors,serum and fecal harmful substances(TMAO,LPS)content,GC-MS detection intestinal contents and monitor the content of short-chain fatty acids(SCFAs)in feces,LC-MS detects the content of bile acid metabolites in intestinal contents,and refers to the signal pathways of differential gene expression and enrichment in midgut transcriptomics,from morphology,biochemical,omics and other multi-angle analysis of the effect of RES on the intestinal lipid synthesis of red tilapia.Results were as follows:1.High-fat feeding will destroy the morphology of the intestinal tissue,and the annular folds of the intestine will be damaged.Adding RES can ensure the structural integrity of the intestine and significantly increase the number of goblet cells in the midgut;2.After high-fat feeding,it will increase the expression of immune factors in the intestinal tissue.After adding RES,the content of immune factors(IL-1β,IL-6,IL-10,TNF-α)can be significantly reduced,and the content of serum and feces hazardous substances(TMAO and LPS)play a significant inhibitory effect.The content of SCFAs in the intestinal contents and feces of red tilapia has no significant effect,but the addition of RES will reduce the content of acetic acid in the intestinal contents;3.In normal fat supply,red tilapia mainly uses bile secreted by the liver to complete the regulation of lipid metabolism;after HFD,red tilapia uses intestinal anaerobic microorganisms to produce bound bile acids to complete the regulation of lipid metabolism;after adding RES,it will improve fat metabolism and maintain the body’s normal metabolic level.RES promotes the body to complete the process of fatty acidβoxidation and arachidonic acid metabolism,while RES reduces the cellular immune response process.The addition of inhibitors will increase the expression of the phagosome transcriptome and reduce the process of fatty acidβoxidation metabolism.After high-fat feeding,red tilapia intestinal structure will be damaged and the expression of inflammatory factors will be increased.RES will reduce the body’s immune response and inhibit the expression of inflammatory factors to ensure the body’s health.RES promotes the secretion of free bile acids and regulates the process of lipolysis,and transcriptomics results also show that the differentially expressed genes in the process of fat metabolism and synthesis are significantly enriched.
